神經(jīng)生長因子對貓雙眼融合功能影響的探討

【摘要】:目的: 從分子水平觀察神經(jīng)生長因子(nerve growth factor, NGF)在不同年齡正常貓融合中樞視皮層17區(qū)的表達(dá)量以探討融合功能的發(fā)生機(jī)制及與其功能相關(guān)的可能因素。 方法: 正常2~3周健康家貓12只,應(yīng)用圖形視覺誘發(fā)電位(pattern visual evoked potential, PVEP)檢測4周、6周、12周、16周幼貓雙眼P波振幅和潛時,比較不同年齡P波及雙眼間P波的差別。應(yīng)用免疫組織化學(xué)方法比較不同年齡正常貓融合中樞視皮層17區(qū)NGF免疫陽性細(xì)胞量的差別,以判斷NGF對雙眼融合功能的影響。 結(jié)果: PVEP檢測結(jié)果顯示正常貓雙眼間P波振幅和潛時之間均無統(tǒng)計學(xué)意義(P0.05),正常貓雙眼間無明顯差別。隨著正常貓年齡的增長,P波振幅呈逐漸增加,潛時呈逐漸減少趨勢。免疫組織化學(xué)方法觀察視皮層17區(qū)陽性表達(dá)發(fā)現(xiàn)從4周齡開始皮層17區(qū)皮質(zhì)Ⅱ?qū)?Ⅳ層NGF免疫陽性細(xì)胞量逐漸增加,主要表現(xiàn)為顆粒細(xì)胞增多,6周時達(dá)到最大,從6周到12周逐漸下降,12周時降至接近但仍高于4周水平,16周降至明顯低于4周時的表達(dá)水平。6周時表達(dá)量與4周時差異有高度統(tǒng)計學(xué)意義(P0.001),6周時與12周、16周NGF表達(dá)量差異均有高度統(tǒng)計學(xué)意義(P0.001),4周時與12周時NGF陽性細(xì)胞表達(dá)量差異有統(tǒng)計學(xué)意義(P0.05),與16周時NGF陽性細(xì)胞表達(dá)量差異有高度統(tǒng)計學(xué)意義(P0.001)。 結(jié)論: 隨著正常貓年齡的增大,P波振幅逐漸增加,潛時逐漸減少。PVEP能夠簡單反映視功能的發(fā)育情況,說明隨著年齡增長,視力也在不斷發(fā)育的。根據(jù)各年齡正常幼貓視皮層17區(qū)Ⅱ?qū)?Ⅳ層NGF表達(dá)量的差異可知,NGF與雙眼融合功能的發(fā)育和形成有關(guān)。貓雙眼融合功能隨年齡增大逐漸發(fā)育,于視覺可塑性關(guān)鍵期6周左右發(fā)育最明顯,之后呈逐漸下降趨勢,NGF表達(dá)的變化與雙眼視覺發(fā)育關(guān)鍵期保持高度一致,說明NGF是融合功能可塑性相關(guān)的重要分子基礎(chǔ)。
[Abstract]:Objective: The expression of nerve growth factor (NGF) in the 17 area of fusion center of normal cats at different ages was observed at molecular level in order to explore the mechanism of fusion function and the possible factors related to its function. Methods: Visual evoked potential (VEP) pattern visual evoked potential, PVEP) was used to detect the amplitude and latent time of P wave in the eyes of young cats at 4 weeks, 6 weeks, 12 weeks and 16 weeks, and the differences of P waves between eyes of different ages were compared. Immunohistochemical method was used to compare the number of NGF immunoreactive cells in the 17 region of the fusion central visual cortex of normal cats of different ages, in order to determine the effect of NGF on binocular fusion function. Results: The results of PVEP showed that there was no significant difference in P wave amplitude and latent time between normal cats and normal cats, but there was no significant difference between normal cats and normal cats. The amplitude of P wave increased gradually and the latent time decreased with the increase of normal cat age.
