Serglycin穩(wěn)定干擾增加鼻咽癌高轉(zhuǎn)移細胞對順鉑的敏感性

【摘要】:目的觀察serglycin穩(wěn)定干擾后對鼻咽癌高轉(zhuǎn)移細胞順鉑敏感性的影響,并初步探究其機制。方法建立serglycin穩(wěn)定干擾的鼻咽癌高轉(zhuǎn)移細胞株,qRT-PCR和Western blot檢測serglycin的表達情況。四甲基偶氮唑藍法測定順鉑對穩(wěn)定株細胞的IC50;增殖曲線實驗觀察在順鉑壓力下細胞增殖的差異;克隆形成實驗觀察細胞克隆形成能力的差異;Annexin V/PI雙染檢測細胞凋亡。qRT-PCR檢測穩(wěn)定株細胞中干性基因的表達情況。結(jié)果 Serglycin穩(wěn)定干擾的鼻咽癌高轉(zhuǎn)移細胞株成功建立,細胞形態(tài)發(fā)生改變。四甲基偶氮唑藍法測得serglycin穩(wěn)定干擾細胞的順鉑IC50顯著低于對照組細胞,增殖抑制率高于對照組,藥物作用下克隆形成能力減弱,流式凋亡比例增高。Serglycin穩(wěn)定干擾后干性基因表達下調(diào)。結(jié)論 Serglycin穩(wěn)定干擾可以增加鼻咽癌高轉(zhuǎn)移細胞對順鉑的敏感性。
[Abstract]:Objective to investigate the effect of serglycin stable interference on cisplatin sensitivity of nasopharyngeal carcinoma (NPC) cells with high metastasis. Methods the expression of serglycin was detected by qRT-PCR and Western blot in nasopharyngeal carcinoma (NPC) cell lines with stable interference of serglycin. The IC50 of cisplatin on stable cell line was determined by tetramethyl azolium blue method, and the difference of cell proliferation under the pressure of cisplatin was observed by the proliferation curve experiment. Clone formation assay was used to observe the difference of cell clone forming ability. Annexin V/PI double staining was used to detect apoptosis. QRT-PCR was used to detect the expression of dry genes in stable cell lines. Results the high metastatic nasopharyngeal carcinoma cell line with stable interference of Serglycin was successfully established and the morphology of the cell was changed. The cisplatin IC50 of serglycin stable interfering cells was significantly lower than that of control cells, and the inhibition rate of proliferation was higher than that of control group. The expression of dry genes was down-regulated after steady interference of apoptosis with increased flow rate. Conclusion stable interference of Serglycin can increase the sensitivity of high metastatic nasopharyngeal carcinoma cells to cisplatin.
