損毀豚鼠一側(cè)耳蝸后水楊酸鈉給藥對對側(cè)耳螺旋神節(jié)GLU、GABA表達(dá)的影響

【摘要】:目的:觀察損毀豚鼠一側(cè)耳蝸后水楊酸鈉給藥對對側(cè)耳螺旋神經(jīng)節(jié)GLU、GABA表達(dá)的影響 方法:出生后體重在250~300g的成年豚鼠64只隨機分成四組,每組16只。A組:正常對照組;B組:單純手術(shù)組;C組:單純水楊酸鈉給藥組300mg/(kg.d) D組:一側(cè)耳蝸搗毀+水楊酸鈉給藥[300mg/(kg.d)]組A組和B組經(jīng)腹腔給與同等計量的生理鹽水。分別在給藥15天及停藥15天后處死動物,制備耳蝸標(biāo)本,通過免疫組織化學(xué)染色檢測對側(cè)耳蝸GABA、GLu在螺旋神經(jīng)節(jié)的表達(dá)。所有動物術(shù)后均經(jīng)后腿肌肉注射青霉素鈉預(yù)防感染(注射劑量為:160萬U/kg/d,1次/天,共給藥1周)。 結(jié)果:(1):給藥15d后,C組(水楊酸鈉給藥組)右側(cè)耳蝸螺旋神經(jīng)節(jié)GLU陽性細(xì)胞明顯高于A組(正常對照組)和B組(P㩳0.001,P㩳0.001);A組和B組比較無統(tǒng)計學(xué)意義。 D組(耳蝸搗毀+水楊酸鈉給給藥)右側(cè)耳蝸螺旋神經(jīng)節(jié)GLU陽性細(xì)胞表達(dá)與A組和B組比較(P㩳0.001,P㩳0.001)有顯著差異;D組與C組比較P㧐0.05,無統(tǒng)計學(xué)意義。 (2):停藥15天后, C組(水楊酸鈉給藥組)右側(cè)耳蝸螺旋神經(jīng)節(jié)GLU陽性細(xì)胞亦明顯高于A組(正常對照組)和B組(P㩳0.05,P㩳0.01);A組和B組比較無統(tǒng)計學(xué)意義。 D組(耳蝸搗毀+水楊酸鈉給給藥)右側(cè)耳蝸螺旋神經(jīng)節(jié)GLU陽性細(xì)胞表達(dá)與A組和B組比較(P㩳0.001,P㩳0.001)有顯著差異;D組與C組比較P㩳0.05,有統(tǒng)計學(xué)意義. (3):給藥15d后,C組(水楊酸鈉給藥組)右側(cè)耳蝸螺旋神經(jīng)節(jié)GABA陽性細(xì)胞明顯低于A組(正常對照組)和B組(P㩳0.05,P㩳0.05);A組和B組比較無統(tǒng)計學(xué)意義。 D組(耳蝸搗毀+水楊酸鈉給給藥)右側(cè)耳蝸螺旋神經(jīng)節(jié)GABA陽性細(xì)胞表達(dá)與A組和B組比較(P㩳0.01,P㩳0.01)有顯著差異;D組與C組比較P㧐0.05,無統(tǒng)計學(xué)意義。 (4):停藥15天后, C組(水楊酸鈉給藥組)右側(cè)耳蝸螺旋神經(jīng)節(jié)GABA陽性細(xì)胞與A組(正常對照組)和B組比較無統(tǒng)計學(xué)意義;A組和B組比較亦無統(tǒng)計學(xué)意義。 D組(耳蝸搗毀+水楊酸鈉給給藥)右側(cè)耳蝸螺旋神經(jīng)節(jié)GABA陽性細(xì)胞表達(dá)與A組和B組比較無顯著差異;D組與C組比較亦無統(tǒng)計學(xué)意義。 結(jié)論:1.一側(cè)耳蝸搗毀+水楊酸鈉給給藥能使對側(cè)耳GLU陽性細(xì)胞表達(dá)明顯上調(diào),停藥15天后這種改變?nèi)匀淮嬖凇?2.一側(cè)耳蝸搗毀+水楊酸鈉給給藥能明顯下調(diào)對側(cè)耳GABA陽性細(xì)胞表達(dá),停藥15天后這種改變消失。
[Abstract]:Objective: to observe the effect of sodium salicylate on the expression of GLU in spiral ganglion of lateral ear after unilateral cochlear lesion in guinea pigs. Methods: Sixty-four adult guinea pigs weighing 250 ~ 300g after birth were randomly divided into four groups. 16 rats in each group: normal control group, group B: simple operation group, group C: sodium salicylate group, 300mg / kg 路d) D group: unilateral cochlea destroyed salicylate [300mg / kg 路d] group A and B were given intraperitoneally the same amount of normal saline. Cochlear specimens were prepared after 15 days of administration and 15 days after withdrawal. The expression of GABAA-GLu in the spiral ganglion of the contralateral cochlea was detected by immunohistochemical staining. All animals were injected penicillin sodium intramuscularly after operation to prevent infection (1.6 million U / kg 路d ~ (-1) 路d ~ (-1) 路d ~ (-1) 路d ~ (-1)) for one week. Results GLU positive cells in right cochlear spiral ganglion in group C (sodium salicylate group) were significantly higher than those in group A (normal control group) and group B (group B). There was no significant difference between group A and group B. The expression of GLU positive cells in the right cochlear spiral ganglion in group D (treated with sodium salicylate) was significantly different from that in group A and group B. There was no significant difference between group D and group C (P < 0.05). GLU positive cells in right cochlear spiral ganglion in group C (sodium salicylate administration group) were significantly higher than those in group A (normal control group) and group B (group B). There was no significant difference between group A and group B. The expression of GLU positive cells in the right cochlear spiral ganglion in group D (treated with sodium salicylate) was significantly different from that in group A and group B. there was a significant difference between group D and group C (P < 0.05). GABA positive cells in right cochlear spiral ganglion in group C (sodium salicylate group) were significantly lower than those in group A (normal control group) and group B (group B). There was no significant difference between group A and group B. The expression of GABA positive cells in the right cochlear spiral ganglion in group D (treated with sodium salicylate) was significantly different from that in group A and group B. There was no significant difference between group D and group C (P < 0.05). GABA positive cells in right cochlear spiral ganglion in group C (sodium salicylate) were not significantly different from those in group A (normal control group) and group B (control group). There was no significant difference in the expression of GABA positive cells in right cochlear spiral ganglion between group D and group A and group B. there was no significant difference between group D and group C. Conclusion 1. Sodium salicylate on one side of the cochlea could significantly up-regulate the expression of GLU positive cells in the contralateral ear, and the change still existed 15 days after withdrawal. 2. Sodium salicylate on one side of the cochlea could significantly down-regulate the expression of GABA positive cells in the contralateral ear, and the change disappeared 15 days after withdrawal.
