間歇低氧小鼠血清氧化應激指標的變化及N-乙酰半胱氨酸的治療作用

【摘要】:目的觀察慢性間歇低氧((chronic intermittent hypoxia,CIH)小鼠血清氧化應激指標的變化情況,并初步探討N-乙酰半胱氨酸(N-acetylcystein,NAC)的治療作用。 方法將50只小鼠隨機分為正常對照組(unhandled control group, UC) 20只,CIH組30只,分別進行間歇低氧3天、1周、2周、3周、4周;另設CIH 4周+NAC組6只。分別采用硫代巴比妥酸法、羥胺法、化學比色法檢測各組小鼠血清丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(Superoxide Dismutase,SOD)、和谷胱甘肽過氧化物(glutathione peroxidase,GSH-PX)表達水平,觀察隨著間歇低氧造模時間延長,各指標的變化情況,以及NAC的治療作用。 結果(1)在間歇低氧造模第3天時,CIH組小鼠血清MDA水平較對照組有所升高,較UC組比較差異有統(tǒng)計學意義,(CIH 8.98±0.50 nmol/mL UC 7.82±0.49 nmol/mL,p0.05)。隨著間歇低氧時間的延長,CIH組小鼠血清MDA水平呈逐漸升高趨勢,各造模時間段兩組之間比較,差異均有統(tǒng)計學意義(p0.05或p0.01)。另外,在間歇低氧造模3天時,CIH組小鼠血清SOD、GSH-PX水平較對照組有所下降,但差異無統(tǒng)計學意義(p0.05);自低氧第1周始,CIH組小鼠血清SOD、GSH-PX分別為83.30±7.32U/mL、211.5±13.39 U/mL,較UC組95.14±8.4 U/mL、233.84.±15.32 U/mL比較差異有統(tǒng)計學意義(p0.05)。隨著間歇低氧時間的延長,CIH組小鼠血清SOD、GSH-PX水平呈逐漸下降趨勢,各造模時間段兩組之間比較,差異均有統(tǒng)計學意義(p0.05或p0.01)。 (2)間歇低氧4周組+NAC組與單獨間歇低氧4周組相比,SOD,GSH-PX升高,MDA降低,差異有統(tǒng)計學意義(p0.05)。但MDA仍高于UC組,SOD、GSH-PX仍低于UC組,差異有統(tǒng)計學意義(P0.05或P0.01)。 結論CIH可導致機體氧化應激水平增加,且隨著低氧時間延長,機體氧化抗氧化失衡越嚴重。NAC治療能夠改善CIH導致的氧化應激反應。
[Abstract]:Objective to observe the changes of serum oxidative stress indexes in chronic intermittent hypoxia intermittent mice and to explore the therapeutic effect of N-acetylcystein Nacetylcysteine (N-acetylcystein). Methods Fifty mice were randomly divided into control group (n = 30) and control group (n = 20). They were treated with intermittent hypoxia for 3 days, 1 week, 2 weeks, 3 weeks and 4 weeks, and CIH for 4 weeks, NAC group, 6 rats, respectively. The serum levels of malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and glutathione peroxidase (GSH-PX) were detected by thiobarbituric acid method, hydroxylamine method and chemical colorimetry respectively. The changes of each index and the therapeutic effect of NAC. Results 1) on the 3rd day after intermittent hypoxia, the serum MDA level of mice in the control group was higher than that in the control group, which was significantly higher than that in the UC group (8.98 鹵0.50 nmol/mL UC 7.82 鹵0.49 nmol / mL). With the prolongation of intermittent hypoxic time, the serum MDA level of mice increased gradually, and there were significant differences between the two groups in each model time period (p0. 05 or p0. 01). In addition, the serum level of GSH-PX in the CIH group was lower than that in the control group after 3 days of intermittent hypoxia, but the difference was not significant (p 0.05), and the serum GSH-PX in the CIH group was 83.30 鹵7.32 U / mL 211.5 鹵13.39 U / mLrespectively since the first week of hypoxia, which was significantly higher than that in the UC group (95.14 鹵8.4 U / mL / L 233.84 鹵15.32 U/mL). The level of GSH-PX in serum of CIH group decreased gradually with the prolongation of intermittent hypoxia time, and there were significant differences between the two groups in each model time period (P 0.05 or p 0.01). (2) compared with intermittent hypoxia for 4 weeks, the GSH-PX level in NAC group was significantly lower than that in intermittent hypoxia group for 4 weeks, and the difference was statistically significant (P 0.05). However, MDA was still higher in UC group than in UC group, and GSH-PX was still lower than that in UC group (P 0.05 or P 0.01). Conclusion CIH can increase the level of oxidative stress, and with the prolongation of hypoxic time, the more serious the imbalance of oxidative oxidation. NAC can improve the oxidative stress response induced by CIH.
