鼻咽癌microRNAs差異表達(dá)譜的鑒定

【摘要】:目的:篩選及驗(yàn)證鼻咽癌中microRNAs(miRs)的差異表達(dá)譜。方法:通過芯片高通量表達(dá)譜分析及大規(guī)模微陣列技術(shù),篩選鼻咽癌與炎癥組織中差異表達(dá)的miRs,并應(yīng)用RT-QPCR方法驗(yàn)證篩選結(jié)果的可信性。結(jié)果:鼻咽癌組織與對(duì)照組織的miRs存在差異表達(dá),其中差異倍數(shù)大于2倍的人屬miRs共有144種。RT-QPCR發(fā)現(xiàn)與炎癥組織相比,癌組織中miRs-34b、miRs-449b、miRs-7-1表達(dá)顯著下調(diào),而miRs-125b、miRs-184、miRs-196b、miRs-205及miRs-24-1表達(dá)上調(diào),結(jié)果與芯片分析結(jié)果相一致。結(jié)論:差異表達(dá)的miRs可能與鼻咽癌的發(fā)生、發(fā)展密切相關(guān),對(duì)鼻咽癌miRs表達(dá)譜的研究可能為鼻咽癌的早期診斷及治療提供有力的靶向依據(jù)。
[Abstract]:Objective: to screen and verify the differential expression profiles of microRN AsmiRsin nasopharyngeal carcinoma (NPC). Methods: the differentially expressed miRsin nasopharyngeal carcinoma (NPC) and inflammatory tissues were screened by high throughput microarray analysis and microarray technique. The RT-QPCR method was used to verify the credibility of the screening results. Results: there was a significant difference in miRs expression between nasopharyngeal carcinoma tissues and control tissues. There were 144 species of miRs in which the difference times were more than 2 times. RT-QPCR revealed that the expression of miRs-34bnmiRs-449brs-7-1 was significantly down-regulated, while miRs-125bnmiRs-184miRs-184miRs-196bnmiRs-205 and miRs-24-1 were up-regulated. The results are consistent with the results of chip analysis. Conclusion: differential expression of miRs may be closely related to the occurrence and development of nasopharyngeal carcinoma. The study of miRs expression profile of nasopharyngeal carcinoma may provide an effective target for the early diagnosis and treatment of nasopharyngeal carcinoma.
