【摘要】：目的探討前炎細(xì)胞因子白介素-17(IL-17)和腫瘤壞死因子-α(TNF-α)對(duì)鼻息肉上皮細(xì)胞中巨噬細(xì)胞炎性蛋白-3α(macrophage inflammatory protein -3α, MIP-3α/CCL20)的作用。 方法離體培養(yǎng)人鼻息肉上皮細(xì)胞,不加刺激因子以及IL-17、TNF-α刺激細(xì)胞后,用酶聯(lián)免疫吸附法(ELISA)測(cè)定上清液中MIP-3α/CCL20含量的變化,并用逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)檢測(cè)人鼻息肉上皮細(xì)胞中MIP-3α/CCL20 mRNA含量的變化。 結(jié)果IL-17、TNF-α單獨(dú)刺激鼻上皮細(xì)胞后,上清液中MIP-3α/CCL20的含量較不加刺激因子時(shí)增多,IL-17、TNF-α共同刺激下,上清液中MIP-3α/CCL20的含量較不加刺激因子、以及IL-17、TNF-α單獨(dú)刺激時(shí)增加。并且IL-17、TNF-α的誘導(dǎo)作用具有劑量-時(shí)間依從性。同時(shí)測(cè)定鼻上皮細(xì)胞中MIP-3α/CCL20 mRNA的相對(duì)表達(dá)量,得到同樣的結(jié)果。 結(jié)論鼻息肉上皮細(xì)胞產(chǎn)生MIP-3α/CCL20,而且IL-17、TNF-α對(duì)其有誘導(dǎo)作用。MIP-3α/CCL20可能對(duì)T細(xì)胞和樹突狀細(xì)胞在鼻息肉的聚集中發(fā)揮重要的作用。
[Abstract]:Objective to investigate the effects of proinflammatory cytokines interleukin-17 (IL-17) and tumor necrosis factor- 偽 (TNF- 偽) on macrophage inflammatory protein -3 偽 macrophage inflammatory protein 3 偽 (MIP-3 偽 / CCL20) in nasal polyp epithelial cells. Methods Human nasal polyp epithelial cells were cultured in vitro. After stimulated by IL-17 TNF- 偽, the levels of MIP-3 偽 / CCL20 in the supernatant were measured by Elisa. The changes of MIP-3 偽 -CCL20 mRNA in human nasal polyp epithelial cells were detected by reverse transcriptase polymerase chain reaction (RT-PCR). Results after stimulation of nasal epithelial cells with IL-17 TNF- 偽 alone, the content of MIP-3 偽 / CCL20 in supernatant was higher than that without IL-17 TNF- 偽, and the content of MIP-3 偽 / CCL20 in supernatant was higher than that of non-stimulator and IL-17 TNF- 偽 alone. Moreover, the induction of IL-17 TNF- 偽 has dose-time compliance. At the same time, the relative expression of MIP-3 偽 / CCL20 mRNA in nasal epithelial cells was measured and the same results were obtained. Conclusion nasal polyp epithelial cells produce MIP-3 偽 / CCL20, and IL-17 TNF- 偽 induces it. MIP-3 偽 / CCL20 may play an important role in the aggregation of nasal polyps by T cells and dendritic cells.
