鋅指轉錄調節(jié)因子KLF4蛋白在喉鱗狀細胞癌組織中的表達及意義

【摘要】: 目的觀察鋅指轉錄調節(jié)因子KLF4(Kruppel-like factor 4)蛋白在人喉鱗狀細胞癌組織中及聲帶息肉組織中的表達的變化。初步分析KLF4在人喉鱗狀細胞癌發(fā)病機制中的作用,為喉鱗狀細胞癌的發(fā)病機制提供實驗依據(jù)。 方法1、免疫組織化學染色經(jīng)病理確診為喉鱗狀細胞癌組織45例(聲門型),觀察KLF4的分布及表達情況,以48例聲帶息肉組織做對照(對照組)。根據(jù)陽性細胞數(shù)的比例和染色強度行半定量積分。2、用Western Bloting法檢測人經(jīng)病理確診12例為喉鱗狀細胞癌(聲門型)組織中KLF4蛋白的含量,以及12例聲帶息肉組織中KLF4的含量。以β-actin為內對照。實驗結果用Image-pro plus Version5.0.1圖像分析系統(tǒng)進行灰度值測定。實驗結果均采用SPSS13.0統(tǒng)計分析軟件進行分析,p0.05為有統(tǒng)計學意義。 結果1、免疫組化:喉鱗狀細胞癌組織中KLF4的陽性表達率為48.9%(22/45)。在聲帶息肉組織中陽性表達率為89.6%(43/48)。兩者間差異具有統(tǒng)計學意義,p 0.05。2、Western bloting:Image-pro plus Version5.0.1圖像分析系統(tǒng)吸光度值,KLF4表達在喉鱗狀細胞癌組織中較聲帶息肉中低,p 0.05,其表達差異有顯著性。 結論1鋅指轉錄調節(jié)蛋白KLF4廣泛表達在聲帶息肉上皮細胞中,主要分布在細胞質及細胞核膜,并且在聲帶息肉組織中的表達比較穩(wěn)定;2、KLF4蛋白在聲門型喉鱗狀細胞癌中的表達低于聲帶息肉組織,兩者間差異具有統(tǒng)計學意義。
[Abstract]:Objective to investigate the expression of zinc finger transcription regulator KLF4(Kruppel-like factor 4 in human laryngeal squamous cell carcinoma and vocal cord polyp.To analyze the role of KLF4 in the pathogenesis of human laryngeal squamous cell carcinoma and to provide experimental evidence for the pathogenesis of laryngeal squamous cell carcinoma.Methods 1. The distribution and expression of KLF4 in 45 cases of squamous cell carcinoma of larynx (glottic type) were observed by immunohistochemical staining, and 48 cases of vocal cord polyp were used as control group (control group).According to the proportion of positive cells and the intensity of staining, the content of KLF4 protein in 12 cases of laryngeal squamous cell carcinoma (glottic type) and the content of KLF4 in 12 cases of vocal cord polyp were detected by Western Bloting method.尾 -actin was used as internal control.The experimental results were measured by Image-pro plus Version5.0.1 image analysis system.The experimental results were analyzed by SPSS13.0 statistical analysis software.Results 1. Immunohistochemistry: the positive expression rate of KLF4 in laryngeal squamous cell carcinoma was 48.9% and 22 / 45%.The positive expression rate was 89.6% in vocal cord polyp tissue.The expression of KLF4 in laryngeal squamous cell carcinoma (LSCC) was significantly lower than that in vocal cord polyp (P 0.05), and there was a significant difference in the expression of KLF4 in laryngeal squamous cell carcinoma (LSCC) than in vocal cord polyp (P 0.05).Conclusion 1 zinc finger transcription regulatory protein (KLF4) is widely expressed in the epithelial cells of vocal cord polyps, mainly in the cytoplasm and nuclear membrane.The expression of KLF4 protein in glottic laryngeal squamous cell carcinoma was significantly lower than that in vocal polyp tissue.
