新型過(guò)氧化物酶體增殖物激活受體激動(dòng)劑對(duì)角膜新生血管的抑制作用

【摘要】:目的:探討過(guò)氧化物酶體增殖物激活受體γ(PPARγ)激動(dòng)劑對(duì)角膜新生血管的抑制作用及機(jī)制。方法:SD大鼠隨機(jī)分成A、B、C及D組,行角膜囊袋法制作角膜新生血管模型,右眼為實(shí)驗(yàn)眼。A組:空白對(duì)照組,正常角膜大鼠;B:陽(yáng)性對(duì)照組,制作模型后生理鹽水滴眼;C、D組:建模后分別滴用0.5%、1.0%PPARγ激動(dòng)劑滴眼液。裂隙燈顯微鏡下測(cè)量新生血管面積,Western blot檢測(cè)角膜血管內(nèi)皮生長(zhǎng)因子(VEGF)表達(dá),ELISA檢測(cè)房水VEGF的表達(dá)變化。結(jié)果:建模后第3,7,14,21,28天,角膜新生血管面積B組為(5.53±0.81)、(14.61±2.78)、(26.12±2.63)、(34.74±3.56)、(37.42±3.87)mm2,C組為(3.75±1.08)、(10.36±1.27)、(18.76±2.35)、(27.88±3.06)、(29.73±3.54)mm2,D組從第7天開(kāi)始為(3.82±0.94)、(8.74±1.06)、(13.93±2.06)、(14.61±2.82)mm2。28dC、D組角膜新生血管抑制率分別為20.6%和61.0%,D組大鼠角膜新生血管面積小于B組和C組,差異有統(tǒng)計(jì)學(xué)意義(P0.05),而C組與B組之間差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。Western blot檢測(cè)燒傷后14d新生血管角膜表達(dá)VEGF明顯增強(qiáng),C、D組VEGF相對(duì)量有不同程度下降。角膜損傷后,大鼠房水中VEGF表達(dá)隨時(shí)間明顯增多,高濃度的PPARγ激動(dòng)劑滴眼液治療后能明顯降低房水VEGF的含量,第21天D組房水VEGF的表達(dá)明顯低于B組[(687±126)pg/ml vs(1 260±104)pg/ml],差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:局部應(yīng)用PPARγ激動(dòng)劑能有效抑制角膜新生血管生成,抑制VEGF的合成和表達(dá)可能是阻止角膜新生血管生長(zhǎng)機(jī)制的一部分。
[Abstract]:Aim: to investigate the inhibitory effect and mechanism of peroxisome proliferator activated receptor (PPAR 緯) agonist on corneal neovascularization.Methods Twenty SD rats were randomly divided into two groups: group C and group D, and the corneal neovascularization model was made with the method of corneal capsule. The right eye was divided into experimental eye (group A): blank control group (group A) and normal cornea group (group B: positive control group).After the establishment of the model, normal saline drops were used in group C: after modeling, 0.5 and 1.0% PPAR 緯 agonist eye drops were used respectively.Measurement of neovascularization area under slit lamp microscope Western blot was used to detect the expression of corneal vascular endothelial growth factor (VEGF).
