HIF-1α和c-met在喉鱗癌中的表達及其與微血管密度的關(guān)系

【摘要】:目的 觀察缺氧誘導(dǎo)因子-1α (hypoxia-inducible factor1alpha, HIF-la)、肝細胞生長因子受體(hepatocyte growth factor receptor, c-met)和微血管密度(microvesel density, MVD)在人喉鱗狀細胞癌的表達及聯(lián)系,探討HIF-1α和c-met在喉鱗狀細胞癌發(fā)生發(fā)展和淋巴轉(zhuǎn)移中的作用,從而為喉癌的發(fā)生機制與基因治療提供一定的理論依據(jù)和實驗基礎(chǔ)。 方法 收集鄭州大學(xué)第一附屬醫(yī)院2006~2010年耳鼻咽喉-頭頸外科所做喉鱗癌標本58例,采用鏈霉菌抗生物素蛋白過氧化物酶(streptavidin peroxidase, SP)法免疫組織化學(xué)技術(shù),檢測HIF-1α和c-met在喉鱗狀細胞癌不同分化類型中的表達,并測定微血管密度(MVD),觀察HIF-1α、c-met和MVD之間的聯(lián)系。 結(jié)果 HIF-1α陽性表現(xiàn)為棕褐色顆粒,主要定位在細胞核,部分在細胞質(zhì)內(nèi)。其在高、中、低分化喉癌中陽性區(qū)平均積分光密度(the average integrated optical density, AIOD)分別為121.91±3.59、133.31±2.65、146.76±6.61,經(jīng)方差分析結(jié)果顯示:不同分化類型間HIF-1α的差異有統(tǒng)計學(xué)意義(P0.05)。在頸淋巴結(jié)轉(zhuǎn)移組與未轉(zhuǎn)移組中陽性區(qū)AIOD分別為142.85±7.48、126.73±5.82,經(jīng)t檢驗結(jié)果顯示:兩組的差異有統(tǒng)計學(xué)意義(P0.05);c-met陽性表現(xiàn)為棕黃色顆粒,主要定位在胞漿及胞膜。其在高、中、低分化喉癌中陽性區(qū)AIOD分別為120.23±3.04、131.04±4.60、159.12±9.26,經(jīng)方差分析結(jié)果顯示:不同分化類型間c-met的差異有統(tǒng)計學(xué)意義(P0.05)。在頸淋巴結(jié)轉(zhuǎn)移組與未轉(zhuǎn)移組中陽性區(qū)AIOD分別為149.88±14.85、124.81±5.87,經(jīng)t檢驗結(jié)果顯示:兩組的差異有統(tǒng)計學(xué)意義(P0.05);高、中、低分化喉癌中MVD分別為11.82±2.38、23.08±4.95、39.44±4.30,經(jīng)方差分析結(jié)果顯示:不同分化類型間MVD的差異有統(tǒng)計學(xué)意義(P0.05)。在頸淋巴結(jié)轉(zhuǎn)移組與未轉(zhuǎn)移組中MVD分別為34.76±7.47、16.36±5.47,經(jīng)t檢驗結(jié)果顯示:兩組的差異有統(tǒng)計學(xué)意義(P0.05)。Pearson相關(guān)性分析方法分析結(jié)果顯示:HIF-1α與c-met之間的相關(guān)系數(shù)r=0.929,P0.001,有統(tǒng)計學(xué)意義,說明HIF-1α與c-met呈明顯正相關(guān);HIF-1α與MVD之間的相關(guān)系數(shù)r=0.967,P0.001,有統(tǒng)計學(xué)意義,說明HIF-1α與MVD也呈明顯正相關(guān);c-met與MVD之間的相關(guān)系數(shù)r=0.921,P0.001,有統(tǒng)計學(xué)意義,說明兩者也呈正相關(guān)。 結(jié)論 HIF-1α、c-met和新生血管的形成在喉鱗狀細胞癌的發(fā)生、發(fā)展、淋巴結(jié)轉(zhuǎn)移中起重要作用,HIF-1α、c-met存在有協(xié)同作用。HIF-1α、c-met可能作為治療喉鱗癌的潛在靶點。
[Abstract]:PurposeTo investigate the expression and association of hypoxia-inducible factor-1 alpha-hypoxia-inducible factor-1 (HIF-lalaa), hepatocyte growth factor receptor (c-metr) and microvesel density (MVD) in human laryngeal squamous cell carcinoma (LSCC), and to explore the role of HIF-1 偽 and c-met in laryngeal squamous cell carcinoma (LSCC) development and lymphatic metastasis.It provides a theoretical and experimental basis for the pathogenesis and gene therapy of laryngeal carcinoma.MethodFifty-eight cases of laryngeal squamous cell carcinoma (SCC) from 2006 to 2010 in the first affiliated Hospital of Zhengzhou University were collected. Streptavidin peroxidase (SP) immunohistochemical technique was used.The expression of HIF-1 偽 and c-met in different differentiation types of laryngeal squamous cell carcinoma (LSCC) was detected, and the microvessel density (MVD) was measured. The relationship between HIF-1 偽 c-met and MVD was observed.ResultThe expression of HIF-1 偽 was brown granules, mainly located in the nucleus and partly in the cytoplasm.The mean integral optical density of the average integrated optical was 121.91 鹵3.59133.31 鹵2.65146.76 鹵6.61 in high, middle and low differentiated laryngeal carcinoma, respectively. The results of ANOVA showed that there were significant differences in HIF-1 偽 among different differentiation types.The AIOD of positive region in cervical lymph node metastasis group and non-metastasis group was 142.85 鹵7.48126.73 鹵5.82, respectively. The results of t test showed that the difference between the two groups was statistically significant (P 0.05). The positive rate of c-met was brown granules, mainly located in cytoplasm and cell membrane.The AIOD in high, middle and low differentiated laryngeal carcinoma was 120.23 鹵3.04131.04 鹵4.60159.12 鹵9.26, respectively. The results of variance analysis showed that the difference of c-met between different differentiation types was statistically significant (P 0.05).The AIOD of positive region in cervical lymph node metastasis group and non-metastasis group were 149.88 鹵14.85124.81 鹵5.87, respectively. T test results showed that the difference between the two groups was statistically significant (P 0.05);The MVD in poorly differentiated laryngeal carcinoma was 11.82 鹵2.38 鹵23.08 鹵4.95 鹵39.44 鹵4.30, respectively. The results of variance analysis showed that there was significant difference in MVD between different differentiation types.There was a significant positive correlation between HIF-1 偽 and c-met and the correlation coefficient between HIF-1 偽 and MVD, which was statistically significant. The correlation coefficient between HIF-1 偽 and MVD was also positively correlated with MVD (r = 0.921, P 0.001), which indicated that there was a positive correlation between HIF-1 偽 and MVD, which indicated that there was a positive correlation between HIF-1 偽 and MVD, which indicated that there was also a positive correlation between HIF-1 偽 and MVD.ConclusionHIF-1 偽 c-met and neovascularization play an important role in the development and development of laryngeal squamous cell carcinoma. HIF-1 偽 c-met may be a potential target for the treatment of laryngeal squamous cell carcinoma.
