新生豚鼠高膽紅素耳蝸核谷氨酸、γ-氨基丁酸含量表達(dá)

【摘要】:目的觀察高膽紅素模型新生豚鼠耳蝸核谷氨酸(glutamicacid,Glu)和γ-氨基丁酸(gamma-aminobutyricacid,GABA)含量的變化,探討Glu和GABA在高膽紅素?fù)p傷模型低位聽(tīng)覺(jué)中樞耳蝸核中的作用和意義。方法選擇正常新生豚鼠30只隨機(jī)分為三組:對(duì)照組、低膽紅素(100μg/g)處理組和高膽紅素(200μg/g)處理組,8h后處死動(dòng)物取材處理標(biāo)本免疫熒光觀察各組耳蝸核Glu和GABA含量,Western blotting(線粒體凋亡相關(guān)蛋白檢測(cè))觀察細(xì)胞質(zhì)細(xì)胞色素C含量變化。結(jié)果與對(duì)照組相比,高濃度和低濃度膽紅素處理組耳蝸核中GABA的含量水平明顯下降,差異有顯著性(p0.05),而Glu的含量水平則明顯上升(p0.01);Western blotting顯示細(xì)胞色素C在膽紅素高低濃度處理組表達(dá)明顯升高。結(jié)論新生豚鼠高膽紅素模型Glu和GABA含量的動(dòng)態(tài)變化反映了神經(jīng)元的活動(dòng)狀況,提示二者在膽紅素耳神經(jīng)毒性中可能起重要作用。細(xì)胞質(zhì)細(xì)胞色素C增高說(shuō)明高膽紅素可能介導(dǎo)神經(jīng)細(xì)胞早期凋亡。
[Abstract]:Objective to observe the changes of glutamic acid glutamate (glutamic acid) and gamma-aminobutyric acid (GABA) in the cochlear nucleus of neonatal guinea pigs with hyperbilirubin model. To investigate the role and significance of Glu and GABA in the central cochlear nucleus of low hearing in the model of hyperbilirubin injury, 30 normal newborn guinea pigs were randomly divided into three groups: control group. Low bilirubin 100 渭 g / g and hyperbilirubin 200 渭 g / g) treatment group for 8 h, then killed the animals to observe the content of Glu and GABA in the cochlear nucleus and the content of cytochrome C in the cytoplasm of each group. The results were compared with those in the control group. The content of GABA in cochlear nucleus of high and low concentration bilirubin group decreased obviously. There was a significant difference between the two groups, and the level of Glu was significantly increased. Western blotting showed that the expression of cytochrome C in bilirubin treatment group was significantly increased. Conclusion the dynamic changes of Glu and GABA levels in neonatal guinea pig model of hyperbilirubin are reflected. The activity of neurons, These results suggest that both of them may play an important role in the ototoxicity of bilirubin. The increase of cytochrome C in cytoplasm suggests that hyperbilirubin may mediate early apoptosis of nerve cells.
