12-脂氧化酶與糖尿病性視網(wǎng)膜病變發(fā)生發(fā)展相關(guān)性的研究

【摘要】: 目的:通過對(duì)比糖尿病性視網(wǎng)膜病變(DR)大鼠視網(wǎng)膜內(nèi)12-脂氧化酶的表達(dá)與正常大鼠的差異,證明12-脂氧化酶與糖尿病性視網(wǎng)膜病變發(fā)生發(fā)展的相關(guān)性。為進(jìn)一步研究DR的治療方法打下理論基礎(chǔ)。 方法:建立鏈脲佐菌素(STZ)誘導(dǎo)糖尿病性視網(wǎng)膜病變大鼠模型,監(jiān)測(cè)血糖和體重變化情況,5個(gè)月后觀察實(shí)驗(yàn)組和對(duì)照組大鼠眼球形態(tài)學(xué)變化,采用RT-PCR的方法測(cè)定大鼠視網(wǎng)膜內(nèi)12-LOmRNA表達(dá)情況。 結(jié)果:實(shí)驗(yàn)組分為糖尿病大鼠血糖高組和糖尿病大鼠血糖恢復(fù)組。糖尿病大鼠血糖高組并發(fā)性白內(nèi)障發(fā)生率達(dá)100%,部分大鼠出現(xiàn)虹膜新生血管、周邊視網(wǎng)膜及視盤前新生血管。正常組大鼠視網(wǎng)膜12-LOmRNA相對(duì)表達(dá)量為1.17±0.18,糖尿病大鼠血糖高組為1.96±0.28,糖尿病大鼠血糖恢復(fù)組為1.64±0.33;實(shí)驗(yàn)組與正常組比較表達(dá)量明顯升高,差異具有顯著性意義,糖尿病大鼠血糖恢復(fù)組與糖尿病大鼠血糖高組比較略有減少,但差異不明顯。 結(jié)論:鏈脲佐菌素可以有效誘導(dǎo)大鼠糖尿病性白內(nèi)障及糖尿病性視網(wǎng)膜病變,造模5個(gè)月大鼠可產(chǎn)生增殖期DR改變。STZ誘導(dǎo)的DR大鼠12-LO表達(dá)明顯升高,且血糖恢復(fù)正常對(duì)表達(dá)無明顯影響。
[Abstract]:Objective: to compare the expression of 12-lipoxygenase in the retina of diabetic retinopathy rats and normal rats. To prove the correlation between 12-lipoxygenase and the occurrence and development of diabetic retinopathy, and to lay a theoretical foundation for the further study of the treatment of Dr. Methods: the diabetic retinopathy model was induced by streptozotocin (STZ), and the changes of blood glucose and body weight were monitored. After 5 months, the morphologic changes of the eyes of the experimental group and the control group were observed. The expression of 12-LOmRNA in rat retina was determined by RT-PCR. Results: the experimental group was divided into two groups: diabetic rats with hyperglycemia and diabetic rats with recovery of blood glucose. The incidence of cataract in diabetic rats was 100%, and iris neovascularization was found in some rats. The relative expression of 12-LOmRNA was 1.17 鹵0.18 in the normal group, 1.96 鹵0.28 in the hyperglycemia group and 1.64 鹵0.33 in the diabetic recovery group. There was significant difference between diabetic rats and diabetic rats, but there was no significant difference between diabetic rats and diabetic rats. Conclusion: streptozotocin can effectively induce diabetic cataract and diabetic retinopathy in rats. The 12-LO expression of Dr rats induced by streptozotocin can be significantly increased after 5 months of modeling. And there was no obvious effect on the expression of blood glucose returning to normal.
