【摘要】：為探明茶黃素 3,3' 雙沒食子酸酯(TFDG)對人晶狀體上皮細(xì)胞SRA01/04(Human lens epithelial cells,HLECs SRA01/04)的保護(hù)作用,采用MTT法建立高濃度葡萄糖(HG)誘導(dǎo)的HLECs損傷模型,同時觀察HLECs細(xì)胞形態(tài)的變化。用不同濃度(4、8、16、64、32、64、128、256、512μmol/L)TFDG處理HLECs損傷細(xì)胞,采用化學(xué)比色法和硝酸還原法測定HLECs內(nèi)一氧化氮(NO)的含量和一氧化氮合酶(NOS)的活性。結(jié)果表明:高濃度葡萄糖可以抑制人晶狀體上皮細(xì)胞生長,用于HLECs損傷模型誘導(dǎo)的最佳葡萄糖濃度為50 mmol/L,最佳誘導(dǎo)時間為24 h;與未經(jīng)處理的模型細(xì)胞相比,經(jīng)濃度為16~64μmol/L TFDG處理的模型細(xì)胞的存活率顯著提高(P0.05);各個濃度的TFDG能極顯著降低模型細(xì)胞內(nèi)NOS的活性,極顯著減少NO的合成(P0.01),且呈濃度依賴性。以上結(jié)果表明,TFDG對高濃度葡萄糖誘導(dǎo)的HLECs細(xì)胞損傷具有較好的保護(hù)作用。
[Abstract]:In order to investigate the protective effect of theaflavin (TFDG) on human lens epithelial cells (SRA01/04(Human lens epithelial cells), the HLECs damage model induced by high concentration glucose was established by MTT method. At the same time, the morphological changes of HLECs cells were observed. HLECs damaged cells were treated with different concentrations of HLECs cells. The content of nitric oxide (no) and the activity of nitric oxide synthase (NOS) in HLECs were determined by chemical colorimetry and nitric acid reduction. The results showed that high concentration of glucose could inhibit the growth of human lens epithelial cells. The optimal glucose concentration for HLECs injury model was 50 mmol / L and the optimal induction time was 24 h. The survival rate of the model cells treated with 16 渭 mol/L TFDG was significantly increased, and the activity of NOS in the model cells was significantly decreased by each concentration of TFDG. The results showed that TFDG could protect HLECs cells induced by high concentration of glucose.
