UHRF1在喉鱗狀細(xì)胞癌組織中的表達(dá)及臨床意義

【摘要】:目的:UHRF1是一類(lèi)與細(xì)胞增殖密切相關(guān)的核蛋白,在人類(lèi)多種腫瘤的發(fā)生發(fā)展過(guò)程中扮演著重要作用。然而,UHRF1與喉鱗癌的關(guān)系未見(jiàn)報(bào)道,因此,我們研究UHRF1在喉鱗癌組織中的表達(dá)情況及其臨床意義,為喉癌的發(fā)生發(fā)展及尋求新的治療靶點(diǎn)提供實(shí)驗(yàn)依據(jù)。 方法: 1運(yùn)用qRT-PCR分別檢測(cè)60例喉癌組織和60例對(duì)應(yīng)的癌旁正常組織中UHRF1mRNA表達(dá)水平; 2運(yùn)用免疫組織化學(xué)法和western blot方法檢測(cè)UHRF1蛋白在60例喉癌及60例對(duì)應(yīng)的癌旁正常組織中的表達(dá)情況,分析其與喉癌臨床病理參數(shù)之間的關(guān)系。 結(jié)果: 1qRT-PCR結(jié)果顯示UHRF1mRNA在喉鱗癌組織中的表達(dá)水平明顯高于癌旁正常組織,表達(dá)差異具有顯著統(tǒng)計(jì)學(xué)意義(P0.001); 2免疫組織化學(xué)及western blot結(jié)果顯示UHRF1蛋白在78.3%(47/60)的喉癌組織高表達(dá),而癌旁正常組織未見(jiàn)UHRF1蛋白表達(dá),表達(dá)差異具有顯著統(tǒng)計(jì)學(xué)意義(P0.001); 3UHRF1蛋白高表達(dá)與患者吸煙(P0.001)、較晚的T分期(P=0.005)及臨床分期(P=0.044)、較差的組織學(xué)分化(P=0.048)具有相關(guān)性,而與患者年齡、性別及有無(wú)頸部淋巴結(jié)轉(zhuǎn)移無(wú)關(guān)(P0.05)。 結(jié)論:UHRF1在喉鱗癌組織中表達(dá)上調(diào),,可能與喉癌的發(fā)生發(fā)展有著密切關(guān)系,其可能成為判斷喉癌生物學(xué)行為的重要指標(biāo)。
[Abstract]:Objective: UHRF1 is a kind of nuclear protein closely related to cell proliferation, which plays an important role in the occurrence and development of various human tumors. However, the relationship between UHRF1 and laryngeal squamous cell carcinoma has not been reported. We studied the expression of UHRF1 in laryngeal squamous cell carcinoma (LSCC) and its clinical significance in order to provide experimental evidence for the occurrence and development of laryngeal carcinoma and seek for new therapeutic targets. Methods:. 1the expression of UHRF1mRNA in 60 cases of laryngeal carcinoma and 60 cases of adjacent normal tissues were detected by qRT-PCR. 2Immunohistochemical method and western blot method were used to detect the expression of UHRF1 protein in 60 cases of laryngeal carcinoma and 60 cases of adjacent normal tissues, and to analyze the relationship between the expression of UHRF1 protein and the clinicopathological parameters of laryngeal carcinoma. Results:. 1qRT-PCR results showed that the expression of UHRF1mRNA in laryngeal squamous cell carcinoma tissues was significantly higher than that in adjacent normal tissues, and the difference was statistically significant (P 0.001). 2the results of immunohistochemistry and western blot showed that the expression of UHRF1 protein was high in the laryngeal cancer tissues of 78.3% and 47 / 60%, but no expression of UHRF1 protein was found in the adjacent normal tissues. The difference was statistically significant (P 0.001). The high expression of 3UHRF1 protein was correlated with smoking, late T stage (P 0.005) and clinical stage (P < 0.044), and poor histological differentiation (P < 0.048), but not with age, sex and cervical lymph node metastasis (P 0.05). Conclusion the upregulation of the expression of WUHRF1 in laryngeal squamous cell carcinoma may be closely related to the occurrence and development of laryngeal carcinoma, and it may be an important index to judge the biological behavior of laryngeal carcinoma.
