氫氧化鋁佐劑對(duì)變應(yīng)性鼻炎小鼠模型的影響

【摘要】:目的:研究氫氧化鋁佐劑對(duì)變應(yīng)性鼻炎(AR)小鼠模型的影響,探討AR小鼠建模的恰當(dāng)方法。方法:采用卵清蛋白(OVA)鼻內(nèi)致敏、鼻內(nèi)激惹(局部干預(yù)組)和腹腔內(nèi)OVA加用氫氧化鋁Al(OH)3佐劑注射致敏、OVA鼻內(nèi)激惹(系統(tǒng)干預(yù)組)2種方法建立BALB/c小鼠AR模型,動(dòng)態(tài)觀察2種模型小鼠打噴嚏數(shù)和撓鼻次數(shù);應(yīng)用蘇木精-伊紅染色形態(tài)學(xué)觀察小鼠鼻黏膜上皮內(nèi)杯狀細(xì)胞和黏膜下腺體增生情況;Luna染色形態(tài)學(xué)觀察2種模型小鼠鼻黏膜內(nèi)嗜酸粒細(xì)胞的浸潤(rùn)并對(duì)其計(jì)數(shù)。以酶聯(lián)免疫吸附試驗(yàn)(ELISA)檢測(cè)2種模型小鼠鼻腔沖洗液和血清中IL-4、IL-5、OVA特異性IgE(sIgE)和IFN-γ的濃度。結(jié)果:局部干預(yù)組小鼠打噴嚏數(shù)和撓鼻次數(shù)顯著高于系統(tǒng)干預(yù)組。蘇木精-伊紅染色觀察前者小鼠鼻黏膜上皮內(nèi)杯狀細(xì)胞和黏膜下腺體增生較后者顯著;Luna染色前者鼻黏膜內(nèi)嗜酸粒細(xì)胞計(jì)數(shù)也顯著高于后者。局部干預(yù)組鼻腔沖洗液和血清中IL-4、IL-5和sIgE的濃度明顯高于系統(tǒng)干預(yù)組,而其IFN-γ的濃度明顯低于系統(tǒng)干預(yù)組。結(jié)論:OVA和Al(OH)3佐劑聯(lián)合經(jīng)腹腔注射致敏可以同時(shí)促進(jìn)Th1和Th2型細(xì)胞免疫反應(yīng),OVA局部鼻內(nèi)致敏、鼻內(nèi)激惹是建立AR小鼠模型的恰當(dāng)方法。
[Abstract]:Objective: to study the effect of aluminum hydroxide adjuvant on allergic rhinitis (ARA) mouse model and to explore the appropriate method of AR mouse model. The AR model of BALB/c mice was established by intranasal irritation (local intervention group) and intraperitoneal injection of intraperitoneal OVA with aluminum hydroxide Al(OH)3 adjuvant (system intervention group). The number of sneezes and the number of nasal scratches were observed dynamically. Hematoxylin-eosin staining morphology was used to observe the infiltration of eosinophilic granulocytes in the nasal mucosa of mice. Enzyme linked immunosorbent assay (Elisa) was used to detect the concentration of IL-4, IL-5, OVA specific IgES IgE) and IFN- 緯 in nasal lavage and serum of two model mice. Results: the number of sneezing and the number of nasal scratches in the local intervention group were significantly higher than those in the systemic intervention group. The number of eosinophil in nasal mucosa of the former mice was significantly higher than that of Luna staining. Nasal lavage fluid and serum in the local intervention group were higher than those in the latter. The concentrations of IL-4 IL-5 and sIgE were significantly higher than those in the systemic intervention group. The concentration of IFN- 緯 was significantly lower than that of the systemic intervention group. Conclusion the intraperitoneal injection of Al(OH)3 adjuvant can promote the local nasal sensitization of Th1 and Th2 type cellular immune response. Nasal irritation is an appropriate method to establish AR mouse model.
