【摘要】：目的探討人類幾丁質(zhì)酶家族Acidic Mammalian Chitinase(AMcase) , Chitotriosidase(CHIT-1), Stabilin-1 Interacting Chitinase-like Protein (SI-CLP) ,Chi3l1(Chitinase 3 like 1),CTBS(Chitobiase)和Oviductal Glycoprotein 1(OVGP-1)在正常鼻粘膜,不伴鼻息肉的慢性鼻-鼻竇炎和伴鼻息肉的慢性鼻-鼻竇炎中的表達。 方法收集10例正常下鼻甲粘膜(inferior turbinate tissues ITTs),9例不伴鼻息肉的慢性鼻-鼻竇炎(chronic rhinosinusitis CRS),9例伴鼻息肉的慢性鼻-鼻竇炎( nasal polyps NPs)患者組織標本,采用實時熒光定量PCR技術(shù)檢測各組中人類幾丁質(zhì)酶家族成員mRNA的表達。 結(jié)果AMCase,CHIT-1,SI-CLP,Chi3l1和OVGP-1在各組中均有表達。CTBS在ITTs組中的陽性率為20%(2?10),CRSsNP中為33%(3?9)和CRSwNPs中為33% (3?9)。與ITTs組相比AMCase與Chi3l1在CRSwNPs組的表達水平明顯上調(diào),分別為2.4倍(P=0.008)和2.16倍(P=0.03)。 在ITTs組中,CHIT-1的表達和SI-CLP (r = 0.76, P = 0.0092),Chi3l1(r = 0.63, P = 0.047),OVGP-1(r = 0.87, P = 0.008)的表達正相關; Chi3l1和OVGP-1的表達(r = 0.67, P = 0.033)正相關。在CRSsNP組,CHIT-1的表達和SI-CLP (r = 0.76, P = 0.015),Chi3l1(r = 0.66, P = 0.049),AMCase(r = 0.73, P = 0.0246),OVGP-1(r = 0.93, P = 0.0002)的表達正相關; AMCase和SI-CLP (r = 0.7 , P = 0.0358)及OVGP-1(r = 0.85, P = 0.0037)的表達正相關。在CRSwNP組中CHIT-1分別和Chi3l1(r = 0.81, P = 0.0072),OVGP-1(r = 0.68, P = 0.042)的表達正相關;Chi3l1和AMCase(r =0.66 , P = 0.049),OVGP-1(r = 0.71, P = 0.00298)的表達正相關; AMCase和OVGP-1(r = 0.71, P = 0.0298)的表達正相關. 結(jié)論幾丁質(zhì)酶家族成員在正常以及炎性狀態(tài)的鼻腔和鼻竇粘膜中有表達,部分家族成員的共表達具有顯著性意義。
[Abstract]:Objective to investigate the expression of Acidic Mammalian Chitinase (Acidic Mammalian Chitinase), Chitotriosidase (CHIT-1), Stabilin-1 Interacting Chitinase-like Protein (SI-CLP) and Oviductal Glycoprotein 1OVGP-1 in normal nasal mucosa, chronic rhinosinusitis without nasal polyps and chronic rhinosinusitis with nasal polyps. Methods the tissue specimens of 9 patients with chronic rhinosinusitis without nasal polyps and 9 patients with nasal polyps sinusitis without nasal polyps were collected. The expression of mRNA in human chitinase family was detected by real-time fluorescence quantitative PCR. Results the positive rate of OVGP-1 in ITTs group was 20? In CRSsNP, there are 33? 9) and CRSwNPs for 33%? Compared with the ITTs group, the expression of AMCase and Chi3l1 were significantly up-regulated in the CRSwNPs group (2.4-fold P0. 008) and 2.16-fold Pmex 0.03 (P < 0.05). In ITTs group, there was a positive correlation between the expression of CHIT-1 and SI-CLP tir = 0.76, P = 0.0092Chi3l1tr = 0.63, P = 0.047OVGP-1tr = 0.87, P = 0.008), and the positive correlation between the expression of Chi3l1 and OVGP-1 was 0.67, P = 0.033). In CRSsNP group, the expression of CHIT-1 was positively correlated with SI-CLP tir = 0.76, P = 0.015, Chi3l1mcr = 0.66, P = 0.0499AMCASE r = 0.66. There was a positive correlation between the expression of AMCase and SI-CLP r = 0. 7, P = 0. 0358) and OVGP-1(r = 0. 85, P = 0. 0037). In the CRSwNP group, CHIT-1 was positively correlated with Chi3l1(r = 0. 81, P = 0. 0072, OVGGP-1, r = 0. 68, P = 0. 042). There was a positive correlation between the expression of AMCase and OVGP-1(r (P = 0.71, P = 0.0298), and the positive correlation between the expression of OVGP-1P = 0.71, P = 0.00298, and the expression of AMCase and OVGP-1(r = 0.71, P = 0.0298). Conclusion chitinase family members are expressed in normal and inflammatory nasal cavity and nasal sinus mucosa, and the coexpression of some family members is significant.
