【摘要】：目的探討表皮生長(zhǎng)因子受體(epidermal growth factor receptor,EGFR)抑制劑抑制大膠質(zhì)細(xì)胞活化對(duì)實(shí)驗(yàn)性急性高眼壓大鼠視網(wǎng)膜神經(jīng)節(jié)細(xì)胞(retinal ganglion cells,RGCs)丟失的影響。方法用健康成年SD大鼠18只,隨機(jī)分為3組:空白對(duì)照組,不做任何處理;實(shí)驗(yàn)組采用前房灌注法制造急性高眼壓模型,造模成功后,按6 mg·kg-1·d-1用量給予大鼠口服EGFR抑制劑AG1478;實(shí)驗(yàn)對(duì)照組造模成功后給予大鼠口服等量生理鹽水。3組均于7 d處死大鼠,觀察各組視網(wǎng)膜結(jié)構(gòu)的變化及采用免疫組織化學(xué)法觀察陽(yáng)性節(jié)細(xì)胞的表達(dá)。結(jié)果通過(guò)免疫組化法發(fā)現(xiàn),隨著造模時(shí)間的延長(zhǎng),視網(wǎng)膜結(jié)構(gòu)變得模糊不清,RGCs陽(yáng)性染色細(xì)胞逐漸減少�？瞻讓�(duì)照組、實(shí)驗(yàn)組、實(shí)驗(yàn)對(duì)照組RGCs Thy-1陽(yáng)性表達(dá)光密度值分別是:143.666 7±4.041 5、139.032 2±1.734 0和101.102 3±2.000 1。實(shí)驗(yàn)組和空白對(duì)照組相比,視網(wǎng)膜Thy-1陽(yáng)性表達(dá)差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),實(shí)驗(yàn)對(duì)照組和空白對(duì)照組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05),實(shí)驗(yàn)組與實(shí)驗(yàn)對(duì)照組視網(wǎng)膜Thy-1陽(yáng)性表達(dá),差異亦有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 EGFR抑制劑通過(guò)抑制大膠質(zhì)細(xì)胞的活化對(duì)實(shí)驗(yàn)性急性高眼壓大鼠模型的RGCs有保護(hù)作用。
[Abstract]:Objective to investigate the effect of epidermal growth factor receptor (EGFR) inhibitor on the loss of retinal ganglion cells and retinal ganglion cells (RGCs) in experimental acute intraocular hypertension rats. Methods 18 adult Sprague-Dawley (SD) rats were used to investigate the effects of epidermal growth factor receptor inhibitor (EGFR) on the loss of retinal ganglion cells (RGCs) in rats with acute intraocular hypertension (AIOP). Three groups were randomly divided into three groups: the blank control group without any treatment, the experimental group using the anterior chamber perfusion method to make the acute intraocular pressure model, after the model was successfully made, Rats were given EGFR inhibitor AG1478 at the dosage of 6 mg 路kg-1 路d-1, and rats in the control group were killed at 7 days after the rats were given the same amount of saline. The changes of retinal structure and the expression of positive ganglion cells were observed by immunohistochemical method. The optical density of RGCs Thy-1 positive expression in the blank control group, experimental group and experimental control group was 143.6667 鹵4.041 5,139.032 2 鹵1.734 0 and 101.102 3 鹵2.000 1 respectively. There was no significant difference in the positive expression of Thy-1 between the experimental group and the control group (P 0.05). There was a significant difference between the experimental control group and the blank control group (P 0.05). The positive expression of Thy-1 in the retina of the experimental group and the experimental control group was positive. Conclusion EGFR inhibitor has protective effect on RGCs of experimental acute intraocular hypertension rat model by inhibiting the activation of macroglia cells.
