【摘要】： 目的：制備SD大鼠眼堿燒傷動(dòng)物模型,行羊膜移植術(shù),觀察P63、增殖細(xì)胞核抗原(proliferating cell nuclear antigen,PCNA)在角膜緣處的表達(dá),進(jìn)一步了解羊膜對(duì)角膜緣干細(xì)胞(LSCs)增殖效應(yīng)的影響。方法：健康SD大鼠40只40眼,實(shí)驗(yàn)組標(biāo)本20只,20眼,行眼堿燒傷加羊膜移植術(shù)；對(duì)照組標(biāo)本20只,20眼,單純行眼堿燒傷；術(shù)后第1、2、3、4周分別取角膜緣組織進(jìn)行固定、石蠟包埋、切片,采用免疫組化技術(shù)檢測(cè)角膜緣組織中P63、PCNA的表達(dá)情況。結(jié)果：羊膜移植組角膜緣組織中P63、PCNA表達(dá)情況顯著高于對(duì)照組,羊膜移植組和對(duì)照組相比較,有統(tǒng)計(jì)學(xué)意義(P0.05)；羊膜移植術(shù)后1周、2周、3周和4周P63、PCNA蛋白在角膜緣干細(xì)胞的表達(dá)不一致。呈線性趨勢(shì),1周達(dá)到高峰,以后逐漸降低。 結(jié)論：眼堿燒傷后行羊膜移植術(shù),對(duì)角膜緣干細(xì)胞有明顯的保護(hù)作用和并可促進(jìn)干細(xì)胞的增殖表達(dá)。
[Abstract]:Objective: to establish an animal model of ocular alkali burn in SD rats, and to observe the expression of P63, proliferating cell nuclear antigen-PCNA in the limbus of cornea after amniotic membrane transplantation. Methods: 40 eyes of healthy SD rats and 20 eyes of experimental group were treated with ocular alkali burn and amniotic membrane transplantation, and 20 eyes of control group were treated with ocular alkali burn and amniotic membrane transplantation. The corneal limbal tissue was fixed, paraffin embedded and sectioned at the 1st week and 2nd week after operation. The expression of PCNA in the limbal tissue was detected by immunohistochemical technique. Results: the expression of PCNA in the limbal tissue of amniotic membrane transplantation group was significantly higher than that in the control group, and the expression of PCNA in the limbal tissue of the amniotic membrane transplantation group was significantly higher than that in the control group. Compared with the control group, the amniotic membrane transplantation group had statistical significance (P 0.05), and the expression of P63 PCNA protein in limbal stem cells was not consistent at 2 weeks, 3 weeks and 4 weeks after amniotic membrane transplantation. The expression of PCNA protein in limbal stem cells reached the peak in a linear trend at 1 week, and decreased gradually after the first week of amniotic membrane transplantation. Conclusion: amniotic membrane transplantation after ocular alkali burn can protect limbal stem cells and promote the proliferation and expression of stem cells.
