骨髓間充質(zhì)干細(xì)胞對視網(wǎng)膜缺血再灌注損傷中Fas和FasL表達(dá)的影響

【摘要】:目的探討骨髓間充質(zhì)干細(xì)胞(bone marrow mesenchymal stem cells,BMSC)移植對大鼠視網(wǎng)膜缺血再灌注損傷(retinal ischemia reperfusion injury,RIRI)凋亡相關(guān)因子Fas和FasL表達(dá)的影響。方法體外貼壁篩選法培養(yǎng)大鼠BMSC。將39只健康成年SD大鼠隨機(jī)分成正常組3只、模型組18只、BMSC移植組18只。各組大鼠均選擇右眼作為實(shí)驗(yàn)眼建立大鼠RIRI模型,正常組不做任何處理。另2組大鼠于RIRI后1 h分別給予玻璃體腔注射PBS緩沖液(模型組)、BMSC(BMSC移植組),并于RIRI后2 h、6 h、12 h、24 h、48 h、72 h各隨機(jī)處死3只SD大鼠,迅速摘取右眼。免疫組織化學(xué)法檢測大鼠視網(wǎng)膜組織Fas和FasL的陽性細(xì)胞數(shù)情況。結(jié)果正常視網(wǎng)膜組織中Fas極少表達(dá),FasL視網(wǎng)膜全層低表達(dá)。RIRI后2 h、6 h、12 h、24 h、48 h、72 h,模型組Fas陽性細(xì)胞數(shù)分別為(20.5±0.9)個(gè)·mm-2、(263.6±15.6)個(gè)·mm-2、(583.5±15.8)個(gè)·mm-2、(1184.6±45.5)個(gè)·mm-2、(684.5±3.5)個(gè)·mm-2、(80.6±4.5)個(gè)·mm-2,BMSC移植組分別為(19.8±0.8)個(gè)·mm-2、(238.5±14.7)個(gè)·mm-2、(473.6±13.6)個(gè)·mm-2、(964.5±23.7)個(gè)·mm-2、(632.6±5.8)個(gè)·mm-2、(72.5±6.3)個(gè)·mm-2;模型組FasL陽性細(xì)胞數(shù)分別為(42.2±0.9)個(gè)·mm-2、(57.3±0.9)個(gè)·mm-2、(365.8±7.5)個(gè)·mm-2、(1004.3±17.9)個(gè)·mm-2、(793.5±8.6)個(gè)·mm-2、(210.6±4.9)個(gè)·mm-2,BMSC移植組分別為(40.5±0.8)個(gè)·mm-2、(55.6±0.7)個(gè)·mm-2、(351.2±6.8)個(gè)·mm-2、(992.8±6.0)個(gè)·mm-2、(683.4±12.9)個(gè)·mm-2、(206.3±4.7)個(gè)·mm-2。與模型組比較,BMSC移植組各時(shí)間點(diǎn)Fas、FasL的陽性細(xì)胞數(shù)明顯減少。Fas在6~48 h各組差異有統(tǒng)計(jì)學(xué)意義(P0.05),FasL在12~48 h各組差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 RIRI后Fas/FasL表達(dá)水平升高,而玻璃體腔內(nèi)注射BMSC抑制Fas/FasL的表達(dá),證明BMSC移植對RIRI有治療作用。
[Abstract]:Objective to investigate the effect of bone marrow mesenchymal stem cells (BMSCs) transplantation on the expression of Fas and FasL, the apoptosis-related factors of retinal ischemia reperfusion injury in rats with retinal ischemia-reperfusion injury. Methods BMSCs were cultured by adherent screening in vitro. Adult SD rats were randomly divided into normal group (n = 3) and control group (n = 3). There were 18 rats in the model group and 18 rats in the BMSC transplantation group. All the rats in each group were selected as experimental eyes to establish RIRI model. The other two groups were given intravitreous injection of PBS buffer solution 1 h after RIRI (model group), and 3 SD rats were randomly killed at 2 h, 6 h, 12 h, 24 h, 48 h and 72 h after RIRI, respectively. The number of Fas and FasL positive cells in rat retina was detected by immunohistochemical method. Results the low expression of Fas in the whole retinal layer of normal retina was rarely detected. 2 h after 2 h, 12 h, 12 h, 24 h and 48 h and 72 h after RIRI, the model was established. The number of FasL positive cells in the BMSC transplantation group was significantly lower than that in the BMSC transplantation group at 648 h after 648 h. The number of FasL was 351.2 鹵6.8. the number of FasL was significantly lower in BMSC transplantation group than that in BMSC transplantation group (P 0.05) at 1248 h. There was a significant difference in the number of Fas-FasL positive cells between the two groups at 1248 h (P 0.05) and P 0.05 (P 0.05) at 1248 h. The number of Fas-FasL positive cells was significantly lower in BMSC transplantation group than that in the model group at each time point (P < 0.05). The number of FasL positive cells in BMSC transplantation group was significantly lower than that in BMSC transplantation group at 648 h. There were significant differences in the number of Fas-FasL positive cells between two groups (P 0.05) at 1248 h. Conclusion the expression of Fas/FasL increased after RIRI. Intravitreal injection of BMSC inhibits the expression of Fas/FasL, indicating that BMSC transplantation has therapeutic effect on RIRI.
