丙丁酚對(duì)高糖環(huán)境下人視網(wǎng)膜血管內(nèi)皮細(xì)胞增殖及VEGF表達(dá)的影響

【摘要】:目的研究不同濃度的丙丁酚(普羅布考,Probucol)對(duì)體外培養(yǎng)的高糖環(huán)境下人視網(wǎng)膜微血管內(nèi)皮細(xì)胞(human retinal capillary endothelial cells,HRCEC)增殖及血管內(nèi)皮細(xì)胞生長因子(vascular endothelial growth factor,VEGF)表達(dá)的影響。方法 體外培養(yǎng)從角膜移植術(shù)后新鮮人眼球提取的HRCEC。取生長良好的第3-4代細(xì)胞用于實(shí)驗(yàn),實(shí)驗(yàn)分為低糖對(duì)照組、高糖對(duì)照組、高糖+不同濃度丙丁酚(5μmol.L-1、10μmol.L-1、20μmol.L-1、40μmol.L-1、80μmol.L-1)組,用噻唑藍(lán)比色法(MTT)檢測(cè)24h不同濃度的丙丁酚對(duì)其增殖的抑制作用。免疫細(xì)胞化學(xué)法檢測(cè)低糖對(duì)照組、高糖對(duì)照組和不同濃度丙丁酚(5μmol.L-1、40μmol.L-1、80μmol.L-1)組HRCEC中VEGF的表達(dá)。結(jié)果 MTT比色法結(jié)果顯示:不同濃度的丙丁酚作用于原代培養(yǎng)HRCEC24h,細(xì)胞增生抑制率分別為8.40%、20.84%、34.90%、45.32%、55.38%。一定質(zhì)量濃度范圍內(nèi)丙丁酚可抑制高糖環(huán)境下HRCEC的增殖(P0.05),24h內(nèi)呈質(zhì)量濃度依賴性。低糖對(duì)照組與高糖對(duì)照組相比差異無統(tǒng)計(jì)學(xué)意義(P=0.0670.05)。低糖對(duì)照組、高糖對(duì)照組與丙丁酚濃度分別為5μmol.L-1、40μmol.L-1、80μmol.L-1作用于HRCEC24h,免疫細(xì)胞化學(xué)背景灰度值與陽性灰度值之差分別為28.75±3.77、71.93±8.37、54.67±4.33、41.43±3.45、29.18±3.37。與高糖對(duì)照組相比,加藥組作用24h VEGF表達(dá)明顯下降,差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05),且呈質(zhì)量濃度依賴性。與低糖對(duì)照組相比,高糖對(duì)照組VEGF的表達(dá)明顯增加(P0.05)。結(jié)論 丙丁酚對(duì)體外培養(yǎng)的高糖環(huán)境下HRCEC的增殖及高糖引起VEGF表達(dá)的上調(diào)具有抑制作用。
[Abstract]:Objective to study the effects of probucol at different concentrations on the proliferation of human retinal capillary endothelial cells and the expression of vascular endothelial growth factor vascular endothelial growth factor (VEGF) in cultured human retinal capillary endothelial cells under high glucose condition. HRCEC extracted from fresh human eyeball after corneal transplantation was cultured in vitro. The experiment was divided into three groups: low glucose control group, high glucose control group and 10 渭 mol 路L -1 propanol 5 渭 mol 路L ~ (-1) and 10 渭 mol 路L ~ (-1) (20 渭 mol 路L ~ (-1)) group. The inhibitory effects of probufol at different concentrations for 24 hours on proliferation were detected by thiazolyl blue colorimetric assay and low glucose control group by immunocytochemistry. The expression of VEGF in HRCEC of high glucose control group and 40 渭 mol 路L ~ (-1) of propofol group (40 渭 mol 路L ~ (-1)) and 80 渭 mol 路L ~ (-1). Results the results of MTT colorimetric assay showed that the inhibition rate of cell proliferation was 8.40% 20.84% (34.90 渭 mol 路L ~ (-1)) and 55.38% in a certain concentration range, respectively, when different concentrations of probufol were applied to the primary culture of HRCEC for 24 h. Propofol inhibited the proliferation of HRCEC in a concentration dependent manner within 24 hours. There was no significant difference between low glucose control group and high glucose control group. The concentration of propofol and propranol were 5 渭 mol 路L ~ (-1) and 40 渭 mol 路L ~ (-1) and 80 渭 mol.L-1, respectively, and the difference between immunocytochemical background gray value and positive gray value was 28.75 鹵3.77 鹵71.93 鹵8.37 鹵54.67 鹵4.33 鹵41.43 鹵3.4529.18 鹵3.37 respectively. The difference was statistically significant (P 0.05) and in a concentration dependent manner. Conclusion Probufol can inhibit the proliferation of HRCEC and the up-regulation of VEGF expression induced by high glucose in vitro.
