鼻敏感顆粒對變應(yīng)性鼻炎小鼠樹突狀細(xì)胞免疫調(diào)節(jié)機(jī)制的研究

【摘要】:目的:研究鼻敏感顆粒對變應(yīng)性鼻炎小鼠樹突狀細(xì)胞(DC)發(fā)揮免疫調(diào)節(jié)作用的具體機(jī)制。方法:分離出小鼠骨髓細(xì)胞,使用rGM-CSF和rIL-4共培養(yǎng)骨髓細(xì)胞,刺激骨髓細(xì)胞向DC轉(zhuǎn)化。然后加入脂多糖和不同濃度的鼻敏感顆粒共刺激未成熟DC,12h后檢測DC中MHCⅡ、CD80、CD86表面分子以及細(xì)胞上清中TNF-α、IL-1β、IL-6和IL-10的表達(dá)。結(jié)果:鼻敏感顆粒能夠顯著抑制成熟DC標(biāo)志性細(xì)胞因子IL-6、TNF-α、IL-10、IL-1β的生成和成熟DC標(biāo)志物MHCⅡ、CD80和CD86的表達(dá)。結(jié)論:鼻敏感顆粒能夠顯著抑制未成熟DC向成熟DC的分化。
[Abstract]:Objective: to study the immunomodulatory mechanism of Bi-sensitive granules on dendritic cells of allergic rhinitis mice.
