【摘要】： 目的： 諸多研究表明,EB病毒膜蛋白LMP-1在鼻咽癌的發(fā)生發(fā)展中起重要作用,然而其具體發(fā)病機(jī)制遠(yuǎn)未闡明。我們前期研究發(fā)現(xiàn)：在鼻咽癌中,LMP-1與Cyclin D1的表達(dá)呈正相關(guān)。令人關(guān)注的是,新近研究發(fā)現(xiàn)LMP-1可以激活信號轉(zhuǎn)導(dǎo)通路Ras/MAPKKK/MAPKK/ERK/Ets-1,激活Ets-1,同時又有研究證實(shí),在鼠源腎細(xì)胞中,Ets-1在靶基因cyclin D1啟動子上有結(jié)合位點(diǎn)(5'-GATCTCGAGCAGGAAGTTC GA-3'),可以激活cyclin D1的啟動子,上調(diào)cyclin D1的表達(dá)。因此,我們推測在鼻咽癌中LMP-1、Ets-1和Cyclin D1三者之間存在關(guān)聯(lián)。本項(xiàng)目擬從臨床組織水平探討LMP-1、Ets-1和Cyclin D1蛋白在鼻咽癌中的相關(guān)性及它們的表達(dá),為進(jìn)一步研究LMP-1介導(dǎo)的致病機(jī)制提供實(shí)驗(yàn)依據(jù)。 方法： 我們采用免疫組織化學(xué)Envision法在鼻咽癌(非角化鱗狀細(xì)胞癌)活檢蠟塊標(biāo)本中檢測LMP-1、Ets-1和Cyclin D1的表達(dá)并分析它們之間的相關(guān)性,其中男性96例,女性47例,平均年齡48.2±14.8歲,年齡范圍16至82歲。 結(jié)果： ●LMP-1、Ets-1和Cyclin D1蛋白在鼻咽癌中的陽性表達(dá)率：143例鼻咽癌病理標(biāo)本的免疫組化評價結(jié)果顯示,LMP-1陽性表達(dá)率81.8%(117/143)；Ets-1陽性表達(dá)率95.1%(136/143)；Cyclin D1陽性表達(dá)率91.6%(136/143),三者在鼻咽癌中均高表達(dá)。 ●LMP-1、Ets-1和Cyclin D1在鼻咽癌中的表達(dá)相關(guān)性：相關(guān)性分析結(jié)果顯示：LMP-1的高表達(dá)與Ets-1和Cyclin D1的表達(dá)呈正相關(guān)(LMP-1與Ets-1,P=0.000,r=0.397；LMP-1與Cyclin D1, P=0.001,r=0.315)；同時Ets-1表達(dá)與Cyclin D1的表達(dá)在統(tǒng)計(jì)上顯著相關(guān)(P=0.004,r=0.368)。 結(jié)論： 在鼻咽癌組織中,LMP-1、Ets-1和Cyclin D1高表達(dá),并且表達(dá)密切相關(guān)。
[Abstract]:Objective:. Many studies have shown that Epstein-Barr virus membrane protein LMP-1 plays an important role in the development of nasopharyngeal carcinoma, but its specific pathogenesis is far from clear. Our previous study found that the expression of LMP-1 and Cyclin D1 was positively correlated in nasopharyngeal carcinoma. Recent studies have found that LMP-1 can activate the signal transduction pathway Rasr / MAPKK / MAPKK / ERK- 1, activate Ets-1. At the same time, it has been confirmed that Ets-1 has a binding site on the target gene cyclin D1 promoter on the target gene cyclin D1 promoter, which can activate the promoter of cyclin and up-regulate the expression of cyclin D1. We speculated that there was a correlation between LMP-1Ets-1 and Cyclin D1 in nasopharyngeal carcinoma. This study aims to investigate the relationship between LMP-1Ets-1 and Cyclin D1 proteins and their expression in nasopharyngeal carcinoma at the clinical tissue level. To provide experimental basis for further study of LMP-1 mediated pathogenesis. Methods:. Immunohistochemical Envision method was used to detect the expression of LMP-1Ets-1 and Cyclin D1 in paraffin specimens of nasopharyngeal carcinoma (non-keratinized squamous cell carcinoma). The correlation between LMP-1Ets-1 and Cyclin D1 was analyzed in 96 males and 47 females with an average age of 48.2 鹵14.8 years. The age range is 16 to 82 years. Results:. The positive expression rate of LMP-1Ets-1 and Cyclin D1 protein in nasopharyngeal carcinoma was evaluated by immunohistochemistry in 143 cases of nasopharyngeal carcinoma. The results showed that the positive expression rate of LMP-1 was 81.8%. The positive rate of Ets-1 was 95.1% 136 / 143Cyclin D1, and all of them were highly expressed in nasopharyngeal carcinoma (NPC). Correlation between the expression of LMP-1Ets-1 and Cyclin D1 in nasopharyngeal carcinoma: the results of correlation analysis showed that the high expression of LMP-1 was positively correlated with the expression of Ets-1 and Cyclin D1, and the expression of LMP-1 was positively correlated with that of Ets-1 and Cyclin D1. The expression of LMP-1 was positively correlated with the expression of Cyclin D1, and the expression of Ets-1 was significantly correlated with the expression of Cyclin D1. Conclusion:. The expression of LMP-1Ets-1 and Cyclin D1 was highly expressed in nasopharyngeal carcinoma (NPC), and the expression was closely related to the expression of LMP-1Ets-1 and Cyclin D1.
