葡萄糖轉(zhuǎn)運(yùn)蛋白1對(duì)人喉鱗狀細(xì)胞癌的影響的初步研究

【摘要】: 目的觀察葡萄糖轉(zhuǎn)運(yùn)蛋白1(glucose transporter 1,GLUT1)在人喉鱗狀細(xì)胞癌中的表達(dá)情況及其與臨床病理學(xué)的關(guān)系;探討GLUT1對(duì)喉癌淋巴轉(zhuǎn)移的影響。 方法應(yīng)用免疫組織化學(xué)的方法以及逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)法檢測(cè)57例喉鱗狀細(xì)胞癌患者的腫瘤組織、癌旁喉組織以及遠(yuǎn)離腫瘤的正常喉組織中GLUT1的蛋白和mRNA的表達(dá),并結(jié)合喉癌患者的臨床病理特征進(jìn)行分析;通過免疫組化方法,用VEGFR-3標(biāo)記淋巴管,分析GLUT1對(duì)喉癌淋巴轉(zhuǎn)移的作用。 結(jié)果(1)GLUT1免疫組化:GLUT1在喉癌組織中的表達(dá)高于癌旁喉組織和正常喉組織(P0.01);在低分化喉癌中的表達(dá)高于高分化喉癌(P=0.043);相比Ⅰ期+Ⅱ期喉癌,Ⅲ期+Ⅳ期喉癌有高表達(dá)GLUT1的趨勢(shì)(P=0.003);有淋巴轉(zhuǎn)移的喉癌的表達(dá)高于無淋巴轉(zhuǎn)移的喉癌(P=0.021);在直徑≥3cm的腫瘤中的表達(dá)高于直徑3cm的腫瘤(P=0.032)。(2)RT-PCR結(jié)果:GLUT1在喉癌組織中的表達(dá)高于癌旁喉組織和正常喉組織(P0.01);在低分化喉癌中的表達(dá)高于中分化和高分化喉癌(P=0.026);相比Ⅰ期+Ⅱ期喉癌,Ⅲ期+Ⅳ期喉癌有高表達(dá)GLUT1的趨勢(shì)(P=0.018);有淋巴轉(zhuǎn)移的喉癌的表達(dá)高于無淋巴轉(zhuǎn)移的喉癌(P=0.04);在直徑≥3 cm的腫瘤中的表達(dá)高于直徑3 cm的腫瘤(P=0.048)。(3)VEGFR-3免疫組化:在喉癌組織中的表達(dá)高于癌旁喉組織和正常喉組織(P0.01);有淋巴轉(zhuǎn)移的喉癌組織的表達(dá)高于無淋巴轉(zhuǎn)移的喉癌(P=0.012)。 結(jié)論GLUT1與喉癌發(fā)生、發(fā)展、分化、淋巴轉(zhuǎn)移、轉(zhuǎn)歸等密切相關(guān),可以作為喉癌的診斷、治療、預(yù)后指標(biāo)之一。
[Abstract]:Objective to investigate the expression of glucose transporter 1glucose transporter 1 (GLUT1) in human laryngeal squamous cell carcinoma (LSCC) and its relationship with clinicopathology. To investigate the effect of GLUT1 on lymphatic metastasis of laryngeal carcinoma. Methods the tumor tissues of 57 patients with laryngeal squamous cell carcinoma were detected by immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR). The expression of GLUT1 protein and mRNA in paracancerous laryngeal tissues and normal laryngeal tissues away from the tumor was analyzed in combination with the clinicopathological features of laryngeal cancer patients. The role of GLUT1 in lymphatic metastasis of laryngeal carcinoma was analyzed by immunohistochemical method and VEGFR-3 labeled lymphatic vessels. Results Immunohistochemical staining of 1% GLUT1 in laryngeal carcinoma was higher than that in adjacent laryngeal tissue and normal laryngeal tissue (P0.01).
