轉(zhuǎn)移抑制基因(MTSS1)、微血管密度(MVD)在鼻咽非角化性鱗癌組織中的表達及其意義

【摘要】:目的本研究采用免疫組織化學(xué)技術(shù)檢測腫瘤轉(zhuǎn)移抑制基因(MTSS1)及微血管密度(MVD)在鼻咽非角化性鱗癌中的表達及二者之間相關(guān)性,探討它們在腫瘤發(fā)生、發(fā)展及其與組織學(xué)類型及分化程度、臨床分期、發(fā)生淋巴結(jié)轉(zhuǎn)移情況等的關(guān)系。 方法采用免疫組織化學(xué)PV-9000二步法,檢測54例鼻咽非角化性鱗癌及25例鼻咽慢性炎癥組織中的MTSS1、CD105標(biāo)記的MVD的表達水平。 結(jié)果MTSS1蛋白在鼻咽非角化性鱗癌組織及鼻咽炎癥組織中陽性表達率分別為70.37%和92.00%,,差異具有顯著性,P=0.03;MTSS1蛋白的表達在不同的TNM分期、組織學(xué)分型和是否發(fā)生淋巴轉(zhuǎn)移組間的陽性表達率具有顯著差異,均P0.05;CD105標(biāo)記的MVD在鼻咽非角化性鱗癌腫瘤周圍組織組織中的微血管計數(shù)(13.74±3.47)顯著高于鼻咽慢性炎癥組織(9.23±3.06),P=0.00,MVD計數(shù)在不同TNM分期、組織學(xué)分型和是否發(fā)生淋巴轉(zhuǎn)移組間的微血管計數(shù)具有顯著差異,均P0.05;MTSS1、MVD二者在不同年齡、性別、吸煙史和原發(fā)部位的鼻咽非角化性鱗癌組別間,表達均無顯著差異,P0.05;鼻咽非角化性鱗癌組織中MTSS1蛋白表達與MVD之間尚不能認(rèn)為具有相關(guān)性,P0.05。 結(jié)論MTSS1蛋白在鼻咽非角化性鱗癌中可能對腫瘤發(fā)展及轉(zhuǎn)移具有抑制作用,其表達降低可能與腫瘤惡性生物學(xué)行為或預(yù)后不良有關(guān);MVD在鼻咽非角化性鱗癌組織中高表達與腫瘤具有更高的侵襲性及預(yù)后較差有關(guān);同時,MTSS1和MVD在鼻咽非角化性鱗癌組織中表達不具有相關(guān)性,可能兩者在鼻咽非角化性鱗癌的發(fā)生、轉(zhuǎn)移過程中是兩個相互獨立因素。
[Abstract]:Objective to investigate the expression of tumor metastasis suppressor gene (MTSS1) and microvessel density (MVD) in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC) by immunohistochemical technique. To investigate their relationship with histologic type and differentiation, clinical stage, lymph node metastasis and so on. Methods Immunohistochemical PV-9000 two-step method was used to detect MTSS1 in 54 cases of nasopharyngeal non-keratinizing squamous cell carcinoma and 25 cases of nasopharyngeal chronic inflammation. The expression level of MVD labeled by CD105. Results the positive expression rates of MTSS1 protein in nasopharyngeal non-keratinized squamous cell carcinoma and nasopharyngeal inflammation were 70.37% and 92.00, respectively. There were significant differences in the expression of MTSS1 protein between different TNM stages, histological classification and lymphatic metastasis group (P 0.05). The microvessel count of CD105 labeled MVD in the tissues surrounding nasopharyngeal non-keratinizing squamous cell carcinoma (NSCC) was 13.74 鹵3.47, significantly higher than that in chronic nasopharyngeal inflammation (9.23 鹵3.06). . There was significant difference in MVD count between different TNM stages, histological classification and lymphatic metastasis group (P 0.05). There was no significant difference in the expression of MTSS1 and MVD between different age, sex, smoking history and primary nasopharyngeal non-keratinizing squamous cell carcinoma (NSCC) group (P 0.05). There is no correlation between the expression of MTSS1 protein and MVD in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC). Conclusion MTSS1 protein may inhibit tumor development and metastasis in nasopharyngeal non-keratinized squamous cell carcinoma, and its lower expression may be related to malignant biological behavior or poor prognosis of the tumor. The high expression of MVD in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC) was associated with higher invasion and poor prognosis. There was no correlation between MTSS1 and MVD expression in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC). The expression of MTSS1 and MVD in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC) may be an independent factor in the metastasis of nasopharyngeal non-keratinized squamous cell carcinoma (NSCC).
