海藻糖對(duì)大鼠蒸發(fā)過強(qiáng)型干眼作用機(jī)制及療效的實(shí)驗(yàn)研究

【摘要】:目的 評(píng)價(jià)海藻糖對(duì)蒸發(fā)過強(qiáng)型干眼大鼠模型的療效,并探討其治療機(jī)制。 方法 將55只SD大鼠(共110只眼)隨機(jī)分為三組:正常組(5只,共10只眼)、對(duì)照組(25只,共50只眼)、實(shí)驗(yàn)組(25只,共50只眼)。其中正常組不給予任何處置,對(duì)照組和實(shí)驗(yàn)組通過機(jī)械性阻止大鼠瞬目,暴露角結(jié)膜24小時(shí)制作蒸發(fā)過強(qiáng)型干眼模型,并與模型建立開始分別給予無菌生理鹽水、100mmol/L海藻糖溶液滴雙眼,日4次,共2周。分別于模型建立即刻、1天、3天、7天、14天檢測淚液基礎(chǔ)分泌試驗(yàn)( SchirmerⅠtest,SⅠt)、淚膜破裂時(shí)間(tear film break-up time,BUT)、熒光素染色(fluorescence staining,FLS),檢查后取雙眼角結(jié)膜組織分別行HE染色及免疫組織化學(xué)染色,進(jìn)行角結(jié)膜組織病理學(xué)檢查并觀察炎癥細(xì)胞因子TNF-α、IL-6的表達(dá)。 結(jié)果 1、SⅠt結(jié)果比較:正常組、對(duì)照組和實(shí)驗(yàn)組三組結(jié)果差異無統(tǒng)計(jì)學(xué)意義(P0.05)。 2、BUT:正常組組內(nèi)比較差異無統(tǒng)計(jì)學(xué)意義(P0.05);實(shí)驗(yàn)組與對(duì)照組7天、14天比較差異有統(tǒng)計(jì)學(xué)意義(P0.05);實(shí)驗(yàn)組與正常組14天比較差異有統(tǒng)計(jì)學(xué)意義(P0.01)。 3、FLS:正常組熒光染色積分為0(P0.05);對(duì)照組與實(shí)驗(yàn)組于模型建立即刻積分均達(dá)到高值,對(duì)照組各觀察期積分差異無統(tǒng)計(jì)學(xué)意義(P0.05);實(shí)驗(yàn)組與對(duì)照組自3天開始積分組間差異有統(tǒng)計(jì)學(xué)意義(P0.01); 4、免疫組織化學(xué)顯示結(jié)果比較:對(duì)照組與實(shí)驗(yàn)組大鼠模型建立即刻角結(jié)膜上皮細(xì)胞中均見大量TNF-α、IL-6表達(dá),隨著治療時(shí)間的延長,對(duì)照組TNF-α、IL-6表達(dá)逐漸減少,實(shí)驗(yàn)組TNF-α、IL-6表達(dá)顯著減少。正常組、對(duì)照組和實(shí)驗(yàn)組三組14天組間差異均有統(tǒng)計(jì)學(xué)意義(P0.01)。 結(jié)論 海藻糖可抑制角結(jié)膜上皮炎癥細(xì)胞因子TNF-α、IL-6的表達(dá),減輕角結(jié)膜表面炎癥反應(yīng),提示海藻糖對(duì)干眼的治療具有一定的應(yīng)用前景。
[Abstract]:Purpose To evaluate the therapeutic effect of trehalose on evaporative dry eye rat model and to explore its therapeutic mechanism. Method Fifty-five SD rats (110 eyes) were randomly divided into three groups: normal group (n = 5), control group (n = 25), control group (n = 25) and experimental group (n = 25). There were 50 eyes in which the normal group was not given any treatment, the control group and the experimental group were mechanically prevented from the blink and exposed to the conjunctiva for 24 hours to make a dry eye model with excessive evaporation.
