【摘要】：目的 :探討白細(xì)胞介素-13(interleukin-13,IL-13)基因單核苷酸多態(tài)性(single-nucleotide polymorphism,SNP)與喉癌遺傳易感性的關(guān)系。方法:通過(guò)病例-對(duì)照研究設(shè)計(jì),以111例經(jīng)病理確診的喉癌患者和340例健康對(duì)照者為研究對(duì)象,選取IL-13基因3’-UTR的SNP位點(diǎn)rs1295685,采用TaqMan探針?lè)椒ㄟM(jìn)行多態(tài)性檢測(cè),應(yīng)用Logistic回歸計(jì)算OR值及其95%CI,比較不同基因型與喉癌發(fā)病風(fēng)險(xiǎn)的關(guān)聯(lián)。結(jié)果:病例組與對(duì)照組中rs1295685(IL-13C870T)位點(diǎn)基因型分布存在統(tǒng)計(jì)學(xué)差異,在調(diào)整年齡、吸煙、飲酒及腫瘤家族史因素后發(fā)現(xiàn),與CC基因型相比,攜帶TT/CT基因型者喉癌發(fā)病風(fēng)險(xiǎn)顯著增高(調(diào)整OR值為2.00,95%CI為1.23~3.27,P值為0.005)。與CC基因型相比,攜帶TT/CT基因型在年齡≤62歲、吸煙、飲酒、無(wú)腫瘤家族史各亞組中表現(xiàn)出更高的喉癌患病風(fēng)險(xiǎn)(OR=2.72,95%CI:1.40~5.28;OR=2.07,95%CI:1.19~3.58;OR=1.96,95%CI:1.03~3.73;OR=2.01,95%CI:1.18~3.41)。吸煙、飲酒均與TT/CT基因型存在協(xié)同作用(OR=6.48,95%CI:2.77~15.14和OR=7.35,95%CI:3.15~17.12)。而基因型分布在腫瘤分期、分級(jí)中未見(jiàn)顯著差異。結(jié)論:IL-13基因rs1295685位點(diǎn)的單核苷酸多態(tài)性與喉癌遺傳易感性有關(guān)聯(lián)。
[Abstract]:Objective: to investigate interleukin-13 (IL-13). IL-13) gene single-nucleotide polymorphism. The relationship between SNPs and genetic susceptibility of laryngeal carcinoma methods: 111 patients with laryngeal carcinoma and 340 healthy controls were studied by a case-control study. The polymorphism of SNP locus rs1295685 of IL-13 gene was detected by TaqMan probe method. The OR and 95 CI were calculated by Logistic regression. Results: the genotype distribution of rs1295685 IL-13C870T locus was significantly different between the case group and the control group. After adjusting for age, smoking, alcohol consumption and family history of cancer, it was found that compared with CC genotype, the risk of laryngeal cancer in patients with TT/CT genotype was significantly higher (adjusted OR = 2.00). 95 CI was 1.23 ~ 3.27 (P = 0.005).
