circ-Sirt1抑制NF-κB核轉(zhuǎn)位的機(jī)制及在血管炎癥中的意義

目的:動(dòng)脈粥樣硬化和血管退行性病變是導(dǎo)致心血管疾病的主要病因。炎癥反應(yīng)是導(dǎo)致動(dòng)脈粥樣硬化和血管退行性病的病理基礎(chǔ)。本研究以SIRT1基因外顯子來(lái)源的環(huán)狀非編碼RNA circ-Sirt1為研究對(duì)象,從整體、細(xì)胞和分子水平,探討circ-Sirt1在血管炎癥應(yīng)答中的作用和分子機(jī)制。方法:利用野生型SD大鼠建立頸動(dòng)脈損傷狹窄模型,采用實(shí)時(shí)熒光定量PCR和Western blot方法評(píng)價(jià)circ-Sirt1與血管炎癥損傷的關(guān)系。利用Rip和RNA pull-down結(jié)合Western blot技術(shù)發(fā)現(xiàn)并驗(yàn)證與circ-Sirt1相互作用的蛋白。從RNA與蛋白質(zhì)互作角度探討circ-Sirt1對(duì)NF-κB p65活化及核轉(zhuǎn)位的影響。結(jié)果:1 circ-Sirt1參與VSMCs表型調(diào)節(jié)1.1 RT-PCR篩選人SIRT1基因產(chǎn)生的環(huán)狀RNA通過(guò)對(duì)環(huán)狀RNA數(shù)據(jù)庫(kù)circBase搜索,發(fā)現(xiàn)人SIRT1基因能產(chǎn)生11種環(huán)狀RNA產(chǎn)物,RT-PCR檢測(cè)11種環(huán)狀RNA在人VSMCs中的表達(dá)情況。結(jié)果顯示,hascirc0093887(circ-Sirt1)表...
