【摘要】：目的探索miR-204在衰老心肌細(xì)胞缺氧后處理中的作用。方法經(jīng)D-半乳糖誘導(dǎo)心肌細(xì)胞衰老后,隨機(jī)分為對(duì)照組(ONC組)、缺氧復(fù)氧組(OAR組)、后處理組(OIPO組)。采用ELISA法檢測(cè)培養(yǎng)基中c Tn-Ι的含量;微板法檢測(cè)LDH的含量;XTT法檢測(cè)細(xì)胞活性;熒光定量PCR法檢測(cè)miR-204的mRNA表達(dá),并確定miR-204與c Tn-Ι、LDH的相關(guān)性。結(jié)果與ONC組相比,OAR、OIPO組c Tn-Ι、LDH的含量均增加(P0.05,P0.01);與OAR組相比,OIPO組c Tn-Ι、LDH的含量無顯著性改變。與ONC組相比,OAR、OIPO組miR-204的表達(dá)均降低(P0.01);相關(guān)性分析顯示miR-204與c Tn-Ι、LDH呈現(xiàn)負(fù)相關(guān)。結(jié)論缺氧后處理對(duì)缺氧復(fù)氧所致的衰老心肌細(xì)胞損傷的保護(hù)作用減弱與miR-204有關(guān)。
[Abstract]:Objective to explore the role of miR-204 in anoxic post-treatment of aging cardiomyocytes. Methods after cardiomyocyte senescence induced by D-galactose, the cardiomyocytes were randomly divided into control group (ONC group), hypoxia-reoxygenation group (OAR group) and post-treatment group (OIPO group). The content of c Tn- I in culture medium was detected by ELISA method, the content of LDH was detected by microplate method, the cell activity was detected by XTT method, and the mRNA expression of miR-204 was detected by fluorescence quantitative PCR, and the correlation between miR-204 and c Tn- I, LDH was determined. Results compared with ONC group, the contents of c Tn- I and LDH in OAR,OIPO group were increased (P0.05, P 0.01), but the contents of c Tn- I and LDH in OIPO group had no significant change compared with OAR group. Compared with ONC group, the expression of miR-204 in OAR,OIPO group was decreased (P 0.01), and correlation analysis showed that miR-204 was negatively correlated with c Tn- I and LDH. Conclusion the weakening of protective effect of hypoxia post-treatment on aging cardiomyocyte injury induced by hypoxia and reoxygenation is related to miR-204.
