生理性缺血訓(xùn)練對動脈粥樣硬化進程中兔血管內(nèi)皮的影響

【摘要】:目的:研究動脈粥樣硬化早期進行生理性缺血訓(xùn)練對斑塊形成的影響。方法:成年新西蘭白兔24只,隨機分為3組,每組8只,分別為對照組、高脂組和訓(xùn)練組。對照組采用普通飲食,其余兩組采用高脂飲食,其中訓(xùn)練組左下肢用止血帶結(jié)扎的方式進行生理性缺血訓(xùn)練3min/次,3次/d,5d/周,另外兩組安靜籠養(yǎng)。實驗周期為4周。結(jié)果:4周實驗后,兔主動脈油紅-O染色結(jié)果顯示,高脂組主動脈血管壁上動脈粥樣硬化斑塊面積占1.84±0.83%,而對照組和訓(xùn)練組卻沒有動脈粥樣硬化斑塊形成。血管橫切面HE染色顯示高脂組兔主動脈內(nèi)膜上已經(jīng)形成泡沫細胞,而對照組和訓(xùn)練組兔的主動脈內(nèi)膜光滑,無泡沫細胞形成。實驗前三組兔血液VEGF含量組間差異無顯著性意義(對照組:8.35±0.37pg/ml,高脂組:8.27±0.55pg/ml,訓(xùn)練組:8.63±0.36pg/ml,P0.05),實驗后三組兔血液VEGF含量均高于實驗前,但組間差異無顯著性意義(對照組:16.93±0.89pg/ml,高脂組:14.39±0.97pg/ml,訓(xùn)練組:14.36±0.73pg/ml,P0.05)。實驗前三組兔血液NO含量組間差異無顯著性意義(對照組:0.888±0.23μmol/L,高脂組:0.421±0.09μmol/L,訓(xùn)練組:0.529±0.134μmol/L,P0.05),實驗后三組兔血液NO含量均高于實驗前,組間差異具有顯著性意義(P0.01),訓(xùn)練組(2.18±0.144μmol/L)和高脂組(1.82±0.078μmol/L)均顯著高于對照組(1.27±0.167μmol/L)。實驗前三組兔血液EPCs含量組間差異無顯著性意義(對照組:7.25±0.86個/每10~5個淋巴細胞,高脂組:7.5±0.6個/每10~5個淋巴細胞,訓(xùn)練組:8.87±0.61個/每10~5個淋巴細胞,P0.05),實驗后訓(xùn)練組EPCs含量(12.75±0.94個/每10~5個淋巴細胞)高于高脂組(8.25±0.73個/每10~5個淋巴細胞)和對照組(8.25±0.73個/每10~5個淋巴細胞)且組間差異具有顯著性意義(P0.01)。結(jié)論:在動脈粥樣硬化形成過程中,生理性缺血訓(xùn)練可以促進機體產(chǎn)生血管內(nèi)皮相關(guān)因子,保護血管內(nèi)皮功能,減緩粥樣硬化斑塊形成速度。
[Abstract]:Objective: to study the effect of physiological ischemia training on plaque formation in early stage of atherosclerosis. Methods: twenty-four adult New Zealand white rabbits were randomly divided into three groups: control group, hyperlipidemia group and training group. Normal diet was used in the control group and high fat diet was used in the other two groups. In the training group, the left lower limbs were treated with tourniquet ligation for physiological ischemia training for 3min/ times, 3 times a day, 5 days a week, and the other two groups were quietly caged. The experiment period was 4 weeks. Results: after 4 weeks of experiment, the oil red-O staining of rabbit aorta showed that the area of atherosclerotic plaque was 1.84 鹵0.83% in the hyperlipidemic group, but there was no atherosclerotic plaque in the control group and training group. The cross-sectional HE staining showed that foam cells had been formed in the aortic intima of the hyperlipidemia group, while the aortic intima of the control group and the training group were smooth and no foam cells were formed. There was no significant difference in blood VEGF content among the three groups before the experiment (control group: 8.35 鹵0.37 PG / ml, high fat group: 8.27 鹵0.55 PG / ml, training group: 8.63 鹵0.36 PG / ml,P0.05). After the experiment, the blood VEGF levels of the three groups were all higher than those before the experiment, but there was no significant difference between the three groups (control group: 16.93 鹵0.89 PG / ml, high fat group: 14.39 鹵0.97pg / ml, training group: 14.36 鹵0.73 PG / ml,P0.05). There was no significant difference among the three groups before the experiment (control group: 0.888 鹵0.23 渭 mol / L, high fat group: 0.421 鹵0.09 渭 mol / L, training group: 0.529 鹵0.134 渭 mol / L, P0.05). After the experiment, the blood NO contents of the three groups were all higher than those before the experiment, and there was a significant difference between the three groups (P0.01). The training group (2.18 鹵0.144 渭 mol / L) and the hyperlipidemia group (1.82 鹵0.078 渭 mol / L) were significantly higher than the control group (1.27 鹵0.167 渭 mol / L). There was no significant difference in blood EPCs content among the three groups before the experiment (control group: 7.25 鹵0.86 / 10 / 5 lymphocytes, hyperlipidemic group: 7.5 鹵0.6 / 10 / 5 lymphocytes, control group: 7.25 鹵0.86 / 10 / 5). Training group: 8.87 鹵0.61 lymphocytes / 5 lymphocytes per 10 days, P0.05). After the experiment, the EPCs content in the training group (12.75 鹵0.94 / 10 / 5 lymphocytes) was higher than that in the hyperlipidemia group (8.25 鹵0.73 / 10 / 5 lymphocytes) and the control group (8.25 鹵0.73 / 10 / 5 lymphocytes). The difference was significant (P0.01). Conclusion: in the process of atherosclerosis, physiological ischemia training can promote the production of vascular endothelial factors, protect vascular endothelial function, and slow down the formation of atherosclerotic plaques.
