【摘要】：代謝組學(xué)是近年來(lái)生命科學(xué)的熱門(mén)研究領(lǐng)域,它通過(guò)分析生物樣品中的代謝物,從機(jī)體水平上揭示疾病代謝紊亂特征。越來(lái)越多的研究證實(shí)將代謝組學(xué)運(yùn)用于臨床診療的可行性,檢測(cè)生物體液的"代謝指紋"可以為疾病預(yù)防、早期診斷、精準(zhǔn)治療、預(yù)后評(píng)估和新藥發(fā)現(xiàn)等提供重要支撐。由此,臨床代謝組學(xué)(clinical metabolomics)應(yīng)運(yùn)而生。臨床代謝組學(xué)是指結(jié)合臨床大樣本,采用代謝組學(xué)和生物信息學(xué)等手段,從整體上描繪內(nèi)源性代謝小分子集合在疾病擾動(dòng)下的穩(wěn)態(tài)失衡及藥物干預(yù)下的轉(zhuǎn)歸機(jī)制,揭示疾病潛在的診療生物標(biāo)志物和可干預(yù)靶標(biāo)的一門(mén)學(xué)科。本文從臨床代謝組學(xué)發(fā)展歷程切入,簡(jiǎn)述了其分析技術(shù)手段和研究?jī)?nèi)容,并以冠心病為例,總結(jié)了臨床代謝組學(xué)在疾病診療中的研究進(jìn)展。
[Abstract]:Metabonomics is a hot research field in life science in recent years. It reveals the characteristics of metabolic disorders in organism level by analyzing metabolites in biological samples. More and more studies have confirmed the feasibility of applying metabonomics to clinical diagnosis and treatment, and the detection of "metabolic fingerprints" in biological fluids can provide important support for disease prevention, early diagnosis, accurate treatment, prognosis evaluation and new drug discovery. Therefore, clinical metabonomics (clinical metabolomics) came into being. Clinical metabonomics is to describe the homeostasis of endogenous metabolic small molecules under the disturbance of disease and the mechanism of outcome under the intervention of drugs by means of metabonomics and bioinformatics combined with large clinical samples. Reveal potential diagnostic and therapeutic biomarkers of disease and a discipline that can target interventions. Based on the development of clinical metabonomics, this paper briefly describes its analytical techniques and research contents, and summarizes the research progress of clinical metabolomics in the diagnosis and treatment of diseases by taking coronary heart disease as an example.
