microRNA-150上調(diào)可改善心肌梗死后心肌纖維化及心功能
[Abstract]:Objective Acute myocardial infarction (AMI) is one of the diseases with the highest morbidity and mortality in the world. Myocardial fibrosis after myocardial infarction is an important mechanism of cardiac insufficiency in patients with acute myocardial infarction. Micro RNA is a non-coding RNA, regulated gene expression, which is closely related to acute myocardial infarction. The purpose of this study was to investigate the expression of microRNA-150 in myocardial tissue of rats with myocardial infarction and to further study the effects of miR-150 on myocardial fibrosis and cardiac function after myocardial infarction. Methods the myocardial infarction model was constructed by ligating the left anterior descending branch of coronary artery in Sprague Dawley rats. The lentivirus overexpression of microRNA-150 was used to up-regulate the expression of miR-150 in myocardial tissue of rats. Furthermore, the expression of microRNA-150 and the content of col1 1 and 偽-SMA messenger RNA were detected by RT-PCR, and the levels of col1 1, 偽-SMA protein were detected by Western Blotting. Cardiac function and myocardial fibrosis were detected by color Doppler echocardiography and Masson staining. The expression of microRNA-150 in myocardial tissue of rats with myocardial infarction and its effect on myocardial fibrosis and cardiac function after myocardial infarction were studied. Results the results showed that the expression of microRNA-150 in the myocardium of myocardial infarction margin was decreased on the 28th day after myocardial infarction (P0.05). In addition, upregulating the expression of miRNA-150 in myocardial tissue of myocardial infarction rats can improve the cardiac function after myocardial infarction. Decrease of collagen volume fraction and expression of col1 1 and SMA in myocardial tissue (P0. 001) conclusion the expression of MicroRNA-150 in myocardial tissue of myocardial infarction marginal area is decreased. Upregulating the expression of microRNA-150 in myocardial tissue can inhibit fibrosis and improve cardiac function after myocardial infarction.
