【摘要】：目的:通過觀察血管內(nèi)皮損傷后內(nèi)膜增生過程中血凝素樣氧化低密度脂蛋白受體-1表達(dá)的變化,探討血凝素樣氧化低密度脂蛋白受體-1(LOX-1)在再狹窄中的作用機(jī)制及纈沙坦對(duì)其作用。方法:建立球囊損傷大鼠主動(dòng)脈內(nèi)皮模型,并將大鼠隨機(jī)分為對(duì)照組、損傷組、藥物組,每組分別于術(shù)后14天、28天取動(dòng)脈組織,行HE染色觀察血管內(nèi)膜增生的過程,逆轉(zhuǎn)錄聚合酶鏈反應(yīng)及免疫組化技術(shù)檢測(cè)主動(dòng)脈組織內(nèi)血凝素樣氧化低密度脂蛋白受體-1的m RNA及蛋白的表達(dá)。結(jié)果:術(shù)后14及28d損傷組大鼠主動(dòng)脈血管內(nèi)膜較對(duì)照組均顯著增厚(P0.05),術(shù)后l4及28d纈沙坦藥物組內(nèi)膜增生程度均輕于損傷組(P0.05),術(shù)后14及28d,損傷組血凝素樣氧化低密度脂蛋白受體-1m RNA及蛋白表達(dá)較對(duì)照組均顯著升高(P0.05),藥物組血凝素樣氧化低密度脂蛋白受體-1m RNA及蛋白表達(dá)較損傷組降低(P0.05)。結(jié)論:大鼠主動(dòng)脈內(nèi)皮損傷后,血管內(nèi)膜增生,血凝素樣氧化低密度脂蛋白受體-1表達(dá)增高;纈沙坦可減少血凝素樣氧化低密度脂蛋白受體-1的表達(dá),減輕損傷血管內(nèi)膜增生的程度。
[Abstract]:Aim: to observe the changes of hemagglutinin like oxidized low density lipoprotein receptor-1 during intimal hyperplasia after vascular endothelial injury. To investigate the mechanism of hemagglutinin-like oxidation of low density lipoprotein receptor-1 (LOX-1) in restenosis and the effect of valsartan on it. Methods: the aortic endothelial model was established in rats with balloon injury. The rats were randomly divided into three groups: control group, injury group and drug group. Arterial tissues were taken from each group on the 14th and 28th days after operation, and the process of intimal hyperplasia was observed by HE staining. Reverse transcriptase polymerase chain reaction (RT-PCR) and immunohistochemical technique were used to detect the expression of m RNA and protein of hemagglutinin like oxidized low density lipoprotein receptor-1 in aorta. Results: the intima of aorta in the injury group was significantly thicker than that in the control group on the 14th and 28th days after operation (P0.05), and the intimal hyperplasia degree of valsartan drug group was lighter than that of the injured group on the 4th and 28th days after operation (P0.05). On the 14th and 28th days after operation, the hemagglutinin-like oxidation in the injured group was lower than that in the control group (P0.05). The expression of lipoprotein receptor-1m RNA and protein was significantly higher than that of control group (P0.05), and the expression of hemagglutinin like oxidized low density lipoprotein receptor-1m RNA and protein in drug group was lower than that in injured group (P0.05). Conclusion: after aortic endothelial injury, vascular intima proliferates, hemagglutinin like oxidized low density lipoprotein receptor-1 expression is increased, valsartan can reduce the expression of hemagglutinin like oxidized low density lipoprotein receptor 1, valsartan can reduce the expression of hemagglutinin like oxidized low density lipoprotein receptor 1. Reduce the degree of intimal hyperplasia of injured vessels.
