ADAM17調(diào)控CGRP對血管緊張素Ⅱ誘導的血管平滑肌細胞增殖的抑制效應

【摘要】:目的闡明腫瘤壞死因子α轉(zhuǎn)化酶(ADAM17)調(diào)控降鈣素基因相關(guān)肽(CGRP)的抗血管平滑肌細胞(VSMC)增殖效應。方法以大鼠源性胸主動脈平滑肌細胞株(A10VSMC)為研究對象,采用RNA干擾抑制ADAM17表達;采用蛋白質(zhì)免疫印跡和實時定量PCR檢測蛋白和mRNA表達水平;采用噻唑藍比色法評估細胞活性。結(jié)果 CGRP顯著抑制血管緊張素Ⅱ介導的VSMC增殖。血管緊張素Ⅱ刺激24 h后,VSMC中ADAM17表達顯著增加。CGRP預處理30 min能顯著降低VSMC中ADAM17表達,而CGRP受體拮抗劑CGRP8-37可抑制或逆轉(zhuǎn)上述效應。ADAM17 RNA干擾可顯著抑制血管緊張素Ⅱ介導的VSMC增殖。結(jié)論降低ADAM17表達可能是CGRP抑制血管緊張素Ⅱ介導的VSMC增殖效應的機制。
