miR-92b-3p靶向肌細(xì)胞增強(qiáng)子2D抑制心肌細(xì)胞肥大

【摘要】:【目的】探討微小RNAmicroRNA-92b-3p(mi R-92b-3p)對(duì)心肌細(xì)胞肥大的調(diào)控作用及其作用靶基因!痉椒ā拷⒀芫o張素Ⅱ(Ang-Ⅱ)誘導(dǎo)的C57BL/6小鼠心肌肥厚模型。通過尾靜脈注射膽固醇修飾的mi R-92b-3p模擬物(agomi R-92b-3p)來增加小鼠心肌中mi R-92b-3p水平,分別設(shè)立3個(gè)實(shí)驗(yàn)組:agomi R-negative control(agomi R-NC)+saline組,agomi RNC+Ang-Ⅱ組,agomi R-92b-3p+Ang-Ⅱ組。用Ang-Ⅱ誘導(dǎo)乳小鼠心肌細(xì)胞建立心肌細(xì)胞肥大模型;雙熒光素酶報(bào)告基因?qū)嶒?yàn)檢測(cè)mi R-92b-3p與潛在靶基因MEF2D 3'端非翻譯區(qū)(3'-UTR)的結(jié)合作用;蛋白印跡法檢測(cè)MEF2D及肥厚相關(guān)蛋白的表達(dá)!窘Y(jié)果】(1)Ang-II誘導(dǎo)的小鼠肥厚心肌和肥大心肌細(xì)胞分別與對(duì)照組中心肌肥厚相關(guān)基因ANP、ACTA1和β-MHC水平比較,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);(2)雙熒光素酶報(bào)告基因?qū)嶒?yàn)結(jié)果提示mi R-92b-3p與MEF2D 3′UTR具有相互結(jié)合作用;mi R-92b-3p可在轉(zhuǎn)錄后水平抑制MEF2D的表達(dá);升高mi R-92b-3p或降低MEF2D水平能一致性地抑制Ang-II誘導(dǎo)的乳小鼠心肌細(xì)胞肥大表型!窘Y(jié)論】MEF2D是mi R-92b-3p的靶基因,并介導(dǎo)了mi R-92b-3p發(fā)揮抑制心肌細(xì)胞肥大的作用。
