【摘要】：目的探討強(qiáng)化劑量阿托伐他汀對(duì)不穩(wěn)定型心絞痛患者經(jīng)皮冠狀動(dòng)脈介入治療(PCI)圍手術(shù)期B7-H3和B7-H4表達(dá)的影響。方法將80例不穩(wěn)定型心絞痛患者隨機(jī)分為常規(guī)劑量組(阿托伐他汀20 mg/d,n=40)和強(qiáng)化劑量組(阿托伐他汀80 mg/d,n=40),分別于PCI術(shù)前和術(shù)后18~24 h收集外周靜脈血,采用ELISA檢測(cè)外周血白細(xì)胞介素4(IL-4)、白細(xì)胞介素10(IL-10)、干擾素γ(IFN-γ)、血清可溶性B7-H3(sB7-H3)、可溶性B7-H4(sB7-H4)水平,用實(shí)時(shí)熒光定量PCR檢測(cè)外周血單核細(xì)胞B7-H3、B7-H4 mRNA相對(duì)表達(dá)量。結(jié)果兩組患者PCI術(shù)后IL-10、sB7-H3、sB7-H4水平和B7-H3、B7-H4的mRNA表達(dá)水平均較術(shù)前升高,其中強(qiáng)化劑量組升高更顯著(P0.05)。相反,兩組患者PCI術(shù)后IL-4、IFN-γ水平均降低,且強(qiáng)化劑量組較常規(guī)劑量組下降更明顯(P0.05)。結(jié)論強(qiáng)化劑量阿托伐他汀可能通過促進(jìn)B7-H3、B7-H4表達(dá),從而降低不穩(wěn)定型心絞痛患者PCI術(shù)后免疫炎癥反應(yīng)。
[Abstract]:Objective to investigate the effect of enhanced dose of Atto vastatin on the expression of B7-H3 and B7-H4 in patients with unstable angina pectoris undergoing percutaneous coronary intervention (PCI) in perioperative period of (PCI). Methods 80 patients with unstable angina pectoris were randomly divided into routine dose group (Atto vastatin 20 mg/d,n=40) and enhanced dose group (Atto vastatin 80 mg/d,n=40). Peripheral venous blood was collected before and after PCI at 1824 hours. The levels of interleukin-4 (IL-4), interleukin-10 (IL-10), interferon 緯 (IFN- 緯), serum soluble B7-H3 (sB7-H3) and soluble B7-H4 (sB7-H4) were detected by ELISA. The relative expression of B7-H3OB7-H4 mRNA in peripheral blood monocytes was detected by real-time fluorescence quantitative PCR. Results the levels of IL-10,sB7-H3,sB7-H4 and mRNA expression of B7-H _ 3 and B _ 7-H _ 4 after PCI in both groups were higher than those before operation, especially in the enhanced dose group (P0.05). On the contrary, the level of IL-4,IFN- 緯 decreased in both groups after PCI, and the level of IL-4,IFN- 緯 in the enhanced dose group was significantly lower than that in the conventional dose group (P0.05). Conclusion the enhanced dose of Atto vastatin may decrease the immune inflammation after PCI in patients with unstable angina pectoris by promoting the expression of B7-H _ 3 and B _ 7-H _ 4.
