分別轉(zhuǎn)染TBX18后乳鼠成纖維細(xì)胞對(duì)心肌細(xì)胞搏動(dòng)頻率的影響

【摘要】:目的用TBX18分別轉(zhuǎn)染成纖維細(xì)胞(CFs)與心肌細(xì)胞(NRVMs),探究轉(zhuǎn)染的成纖維細(xì)胞對(duì)整個(gè)共培養(yǎng)系統(tǒng)的影響。方法用TBX18分別轉(zhuǎn)染乳鼠心臟CFs與NRVMs,并分成5組:NRVMs組,TBX18-NRVMs組,TBX18-NRVMs+CFs組,TBX18-NRVMs+GFP(綠色熒光蛋白)-CFs組與TBX18-NRVMs+TBX18-CFs組。培養(yǎng)7d后觀察并比較各組細(xì)胞的搏動(dòng)頻率。結(jié)果共培養(yǎng)7d后發(fā)現(xiàn)TBX18可顯著提高心肌細(xì)胞的自發(fā)搏動(dòng)[(94.20±7.38)b/min vs.(62.10±11.67)b/min,P0.01],且相比TBX18-NRVMs組,GFP-CFs或CFs與TBX18-NRVMs共培養(yǎng)并沒(méi)有明顯改變搏動(dòng)頻率,但CFs具有減緩心肌自發(fā)搏動(dòng)的趨勢(shì),經(jīng)TBX18轉(zhuǎn)染后的CFs與TBX18-NRVMs共培養(yǎng)可顯著提高心肌細(xì)胞的搏動(dòng)頻率[(118.50±7.25)b/min vs.(82.80±15.82)b/min,P0.01]。結(jié)論在分別轉(zhuǎn)染TBX18的成纖維細(xì)胞與心肌細(xì)胞共培養(yǎng)系統(tǒng)中,成纖維細(xì)胞可顯著提高心肌細(xì)胞的搏動(dòng)頻率。
[Abstract]:Objective to investigate the effect of TBX18 on the whole co-culture system by transfection of fibroblasts (CFS) and cardiomyocytes (NRVMs1) respectively. Methods CFS and NRVMswere transfected with TBX18, respectively, and divided into 5 groups: TBX18-NRVMs (TBX18-NRVMs CFS) and TBX18-NRVMs (TBX18-NRVMs GFPs) and TBX18-NRVMs TBX18-CFS (TBX18-NRVMs). After 7 days of culture, the pulsatile frequency of each group was observed and compared. Results after 7 days of co-culture, it was found that TBX18 could significantly increase the spontaneous pulsation of cardiomyocytes [94.20 鹵11.67 7.38)b/min vs.(62.10 鹵11.67 b / min P0.01]. Compared with TBX18-NRVMs, TBX18-NRVMs co-cultured with GFP-CFs or CFS did not significantly change the pulsatile frequency, but CFS had a tendency to slow down myocardial spontaneous pulsation. The co-culture of CFS and TBX18-NRVMs transfected with TBX18 significantly increased the pulsatile frequency of cardiomyocytes [118.50 鹵7.25)b/min vs.(82.80 鹵15.82mb / min P0.01]. Conclusion in the co-culture system of fibroblasts transfected with TBX18, fibroblasts can significantly increase the pulsatile frequency of cardiomyocytes.
