間歇運(yùn)動(dòng)上調(diào)CTRP3表達(dá)抑制心梗大鼠心肌細(xì)胞凋亡及其機(jī)制

【摘要】:探討間歇運(yùn)動(dòng)對(duì)心肌梗死(myocardial infarction,MI)大鼠C1q-腫瘤壞死因子相關(guān)蛋白3(CTRP3)表征及心肌細(xì)胞凋亡的影響。3月齡雄性SD大鼠30只,隨機(jī)選取8只為假手術(shù)對(duì)照組(sham,S),S組只開(kāi)胸穿線不結(jié)扎;余下大鼠永久性結(jié)扎左冠狀動(dòng)脈前降支,術(shù)后存活16只,隨機(jī)分為心梗安靜組(MI)和心梗+間歇運(yùn)動(dòng)組(ME),每組8只。ME組大鼠于術(shù)后1周進(jìn)行為期8周的間歇運(yùn)動(dòng)訓(xùn)練,訓(xùn)練結(jié)束次日,腹腔麻醉取材。RT-qPCR檢測(cè)CTRP3mRNA水平,Western Blot檢測(cè)心肌CTRP3、p-AKT、AKT、p-mTOR、mTOR、細(xì)胞色素C、Bcl-2、Bax、BNP蛋白表達(dá),TUNEL法檢測(cè)心肌細(xì)胞凋亡,Masson染色分析心肌膠原容積(collagen volume fraction,CVF)百分比,血流動(dòng)力學(xué)方法評(píng)定心功能。研究發(fā)現(xiàn):心梗后大鼠心功能顯著降低,心肌纖維化水平顯著增加,CTRP3mRNA表達(dá)顯著升高,蛋白表達(dá)顯著降低,AKT和mTOR蛋白磷酸化上調(diào),細(xì)胞色素C表達(dá)顯著升高,Bcl-2/Bax比值降低,TUNEL陽(yáng)性顆粒增加,BNP蛋白表達(dá)上調(diào)。間歇運(yùn)動(dòng)可顯著改善心梗大鼠心功能,降低心肌纖維化水平,CTRP3基因與蛋白表達(dá)都顯著升高,進(jìn)一步上調(diào)AKT和mTOR蛋白磷酸化,降低細(xì)胞色素C蛋白表達(dá),升高Bcl-2/Bax比值,減少TUNEL陽(yáng)性顆粒,下調(diào)BNP的蛋白表達(dá)。間歇運(yùn)動(dòng)可升高心梗大鼠心肌中CTRP3水平,激活A(yù)KT/mTOR通路,抑制心肌細(xì)胞凋亡,改善心梗心臟病理性重塑,提高心功能。
[Abstract]:Effects of intermittent exercise on the characterization of C1q-TNF- related protein 3CTRP3) and the effect of intermittent exercise on cardiomyocyte apoptosis in rats with myocardial infarction, 30 male Sprague-Dawley (SD) rats aged 3 months were randomly selected as the sham operation control group (n = 8). Left anterior descending coronary artery was permanently ligated and 16 rats survived after operation. They were randomly divided into silent myocardial infarction group and intermittent myocardial infarction exercise group. Eight rats in each group received intermittent exercise training for 8 weeks one week after operation. The next day after training, CTRP3 mRNA level was detected by RT-PCR and CTRP3 mRNA level was detected by Western blot. The expression of cytochrome Bcl-2Bax-BNP protein was detected by Tunel method. Myocardial collagen volume and volume fractionation CVFpercentage were detected by Masson staining, and the cardiac function was evaluated by hemodynamic method. The results showed that after myocardial infarction, cardiac function was significantly decreased, myocardial fibrosis level was significantly increased, CTRP3 mRNA expression was significantly increased, and protein expression was significantly decreased in AKT and mTOR protein phosphorylation. The expression of cytochrome C increased significantly and the ratio of Bcl-2 / Bax decreased. Intermittent exercise significantly improved cardiac function, decreased the level of myocardial fibrosis and increased the expression of CTRP3 gene and protein, further up-regulated the phosphorylation of AKT and mTOR protein, decreased the expression of cytochrome C protein, and increased the ratio of Bcl-2 / Bax. Decreased Tunel positive granules and down-regulated BNP protein expression. Intermittent exercise could increase the level of CTRP3, activate AKT / mTOR pathway, inhibit myocardial cell apoptosis, improve the rational remodeling of myocardial infarction and improve cardiac function in myocardial infarction rats.
