非諾貝特對(duì)兔動(dòng)脈粥樣硬化斑塊形成的影響及機(jī)制

【摘要】:目的探討過氧化物酶體增殖物激活受體家族(PPARs)激動(dòng)劑非諾貝特對(duì)動(dòng)脈粥樣硬化形成的影響及作用機(jī)制。方法將25只雄性新西蘭白兔隨機(jī)分為五組,每組5只,A組予標(biāo)準(zhǔn)飼料喂養(yǎng),其余四組予高脂飼料喂養(yǎng),均喂養(yǎng)8周。C、D、E組喂養(yǎng)4周結(jié)束時(shí)均行經(jīng)股動(dòng)脈球囊損傷術(shù),并分別于手術(shù)當(dāng)天給予生理鹽水、非諾貝特、WY14643(非諾貝特酸的人工合成物)連續(xù)4周。喂養(yǎng)8周結(jié)束時(shí),五組均進(jìn)行股動(dòng)脈內(nèi)膜剝脫術(shù),取股動(dòng)脈,并留取血清。取各組股動(dòng)脈標(biāo)本,HE染色后顯微鏡下觀察血管形態(tài),計(jì)算血管狹窄率;采用免疫組化SP法及Western blotting法檢測股動(dòng)脈PPARα蛋白表達(dá);采用RT-PCR法檢測股動(dòng)脈PPARαmRNA表達(dá);采用ELISA法檢測血清TNF-α、IL-10和P-選擇素水平。結(jié)果 C、D、E組股動(dòng)脈均有動(dòng)脈硬化表現(xiàn),D、E組表現(xiàn)均較C組輕,且E組較D組輕。與A組比較,其余四組管腔狹窄率均升高(P0.05或0.01);與C組比較,D、E組管腔狹窄率降低(P均0.05)。D、E組股動(dòng)脈PPARα蛋白和mRNA表達(dá)均高于C組,血清IL-10水平高于C組,血清TNF-α和P-選擇素水平均低于C組;組間比較P0.05或0.01。結(jié)論非諾貝特可減輕由高脂飼料和球囊損傷引起的兔動(dòng)脈粥樣硬化病變程度,增加IL-10表達(dá)、減少TNF-α和P-選擇素表達(dá),抑制炎癥反應(yīng)可能是其作用機(jī)制。
[Abstract]:Objective to investigate the effect and mechanism of fenofibrate, a peroxisome proliferator activated receptor family (PPARs) agonist, on atherosclerosis. Methods Twenty-five male New Zealand white rabbits were randomly divided into five groups. Five rabbits in each group were fed with standard diet and the other four groups were fed with high-fat diet. Normal saline and fenofibrate WY 14643 (artificial synthesis of fenofibrate) were given on the day of operation for 4 weeks. At the end of 8 weeks, the five groups were treated with femoral artery endarterectomy, femoral artery and serum. The vascular morphology was observed under microscope after HE staining, the expression of PPAR 偽 protein in femoral artery was detected by immunohistochemical SP method and Western blotting method, and the expression of PPAR 偽 mRNA in femoral artery was detected by RT-PCR. Serum levels of TNF- 偽 IL-10 and P- selectin were detected by Elisa. Results Arteriosclerosis was found in all femoral arteries in group C and group E was lighter than that in group D. Compared with group A, the rate of stenosis in the other four groups was significantly higher than that in group A (P0.05 or 0.01%), and the expression of PPAR 偽 protein and mRNA in femoral artery of group C was significantly higher than that in group C (P < 0.05), and the level of IL-10 in serum was higher than that in group C (P < 0.05), and the level of IL-10 in group E was higher than that in group C (P < 0.05). The serum levels of TNF- 偽 and P- selectin were lower than those of group C (P0.05 or 0.01). Conclusion fenofibrate can reduce the degree of atherosclerosis induced by high fat diet and balloon injury, increase the expression of IL-10, decrease the expression of TNF- 偽 and P- selectin, and inhibit the inflammatory reaction.
