【摘要】：目的抵抗素可通過導(dǎo)致內(nèi)皮細(xì)胞功能的失調(diào),參與動(dòng)脈粥樣硬化的發(fā)生和發(fā)展,然而其作用機(jī)制研究較少。文中旨在研究抵抗素對(duì)冠狀動(dòng)脈內(nèi)皮細(xì)胞功能的影響,探討影響抵抗素作用的可能機(jī)制。方法構(gòu)建針對(duì)人血管擴(kuò)張刺激磷蛋白(VASP)的慢病毒表達(dá)載體的VASP mRNA的shRNA干擾序列(LV-si VASP組)和陰性對(duì)照載體(LV-sicntr組),并將其感染高表達(dá)VASP的人冠狀動(dòng)脈內(nèi)皮細(xì)胞,RT-PCR檢測(cè)VASP mRNA的表達(dá),Western blot測(cè)定蛋白的表達(dá)。用不同濃度的抵抗素(0、10、50、100、200 ng/m L)體外作用于人冠狀動(dòng)脈內(nèi)皮細(xì)胞系(HCAECs),MTT法檢測(cè)抵抗素對(duì)細(xì)胞增殖的作用,Transwell小室實(shí)驗(yàn)檢測(cè)細(xì)胞遷移能力的改變。在相同抵抗素濃度100 ng/m L下,檢測(cè)2組對(duì)內(nèi)皮細(xì)胞增殖、遷移影響。免疫熒光法分析2組細(xì)胞表面受體血管內(nèi)皮生長(zhǎng)因子受體2(VEGFR2)表達(dá)情況。采用pull-down法分析2組細(xì)胞Rho A活性。結(jié)果LV-si VASP組RNA表達(dá)抑制率[(68.1±0.8)%]、VASP蛋白表達(dá)抑制率[(59.3±1.7)%]較LV-sicntr組(100%)明顯降低(P0.05)。與0 ng/m L比較,50~200 ng/m L抵抗素作用48 h后,可以顯著促進(jìn)細(xì)胞的增殖(P0.05)。50~200 ng/m L抵抗素處理24 h后,HCAEC呈劑量依賴性促進(jìn)細(xì)胞遷移(P0.05)。沉默VASP后,抑制了抵抗素對(duì)細(xì)胞增殖和遷移的作用(P0.05)。熒光顯微鏡觀察結(jié)果顯示,與LV-sicntr組比較,LV-si VASP組VEGFR2表達(dá)未見明顯變化,而LV-si VASP組Rho A活性較LV-sicntr組減弱[(41.3±3.1)%vs 100%],差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論抵抗素呈濃度依賴性的促進(jìn)冠狀動(dòng)脈內(nèi)皮細(xì)胞增殖、遷移,敲除VASP對(duì)抵抗素功能的影響與減弱的Rho A活性有關(guān)。
[Abstract]:Objective resistin is involved in the occurrence and development of atherosclerosis by causing dysfunction of endothelial cells. The purpose of this study was to investigate the effects of resistin on the function of coronary endothelial cells and the possible mechanism of affecting resistin. Methods the shRNA interference sequence of VASP mRNA targeting human vasodilation-stimulated phosphoprotein (VASP) lentivirus expression vector was constructed in LV-si VASP group and negative control vector LV-sicntr. RT-PCR was used to detect the infection of human coronary artery endothelial cells with high expression of VASP. The expression of VASP mRNA was detected by Western blot. The effect of resistin on proliferation of human coronary artery endothelial cell line (HCAECs) was detected by using different concentrations of resistin in vitro. The effect of resistin on cell proliferation was detected by Transwell chamber assay. Under the same resistin concentration of 100 ng/m L, the effects of two groups on endothelial cell proliferation and migration were detected. The expression of vascular endothelial growth factor receptor 2 (VEGFR 2) was analyzed by immunofluorescence. The activity of Rho A was analyzed by pull-down assay. Results the inhibitory rate of RNA expression in LV-si VASP group [68.1 鹵0.8%] was significantly lower than that in LV-sicntr group (59.3 鹵1.7%). Compared with 0 ng/m L, resistin could significantly promote cell proliferation after 48 h treatment with resistin 50 ng/m L in a dose-dependent manner, and promote cell migration in a dose-dependent manner. Silencing VASP inhibited the effect of resistin on cell proliferation and migration. The results of fluorescence microscope showed that there was no significant change in VEGFR2 expression in LV-si VASP group compared with LV-sicntr group, but the activity of Rho A in LV-si VASP group was lower than that in LV-sicntr group [41.3 鹵3.1)%vs 100%], the difference was statistically significant (P 0.05). Conclusion resistin promotes the proliferation and migration of coronary endothelial cells in a concentration-dependent manner. The effect of knockout of VASP on resistin function is related to the decreased activity of Rho A.
