【摘要】:目的:研究TRPV1在高同型半胱氨酸促進(jìn)內(nèi)皮細(xì)胞凋亡中的作用,闡明辣椒素抗動(dòng)脈粥樣硬化的可能機(jī)制。方法:以人臍靜脈內(nèi)皮細(xì)胞為離體研究對(duì)象,不同濃度的同型半胱氨酸為干預(yù)措施(0mmol/L、0.05mmol/L、0.1mmol/L、0.2mmol/L、0.5mmol/L、1mmol/L、2mmol/L、4mmol/L)及不同時(shí)間(6h、12h、24h、36h、48h)干預(yù)后用CCK-8檢測(cè)細(xì)胞增殖及活性情況,選擇合適的同型半胱氨酸干預(yù)濃度及干預(yù)時(shí)間進(jìn)一步分組并給予辣椒素干預(yù),然后用細(xì)胞免疫熒光檢測(cè)TRPV1蛋白是否在人血管內(nèi)皮細(xì)胞上表達(dá);Western blotting檢測(cè)各組TRPV1蛋白的表達(dá)、TUNEL染色了解各組細(xì)胞凋亡情況。結(jié)果:(1)同型半胱氨酸對(duì)HUVECs的增殖能力有明顯的抑制作用,吸光值從(1.03±0.12)漸下降至(0.42±0.05),且呈明顯的劑量效應(yīng)關(guān)系(P0.01);當(dāng)干預(yù)濃度從0.5mmol/L開(kāi)始、干預(yù)24小時(shí)后對(duì)人臍靜脈內(nèi)皮細(xì)胞增殖具有抑制作用(P0.05);(2)細(xì)胞免疫熒光檢測(cè)在HUVECs上有TRPV1蛋白表達(dá);(3)同型半胱氨酸干預(yù)后TPRV1在0.5mmol/L、1mmol/L、2mmol/L組表達(dá)較對(duì)照組下降(p0.05)、細(xì)胞凋亡較對(duì)照組增加(P0.05);(4)辣椒素干預(yù)后,與單純同型半胱氨酸干預(yù)組比較,TRPV1蛋白表達(dá)升高(p0.05)、細(xì)胞凋亡率下降,且有統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論:高同型半胱氨酸可通過(guò)下調(diào)TRPV1蛋白的表達(dá),從而促進(jìn)HUVECs凋亡;辣椒素可抑制高同型半胱氨酸誘導(dǎo)的內(nèi)皮細(xì)胞凋亡。
[Abstract]:Aim: to investigate the role of TRPV1 in promoting apoptosis of endothelial cells induced by homocysteine, and to elucidate the possible mechanism of capsaicin against atherosclerosis. Methods: human umbilical vein endothelial cells (HUVECs) were treated with different concentrations of homocysteine at 0 mmol / L 0. 05 mmol / L 0. 1 mmol / L 0. 2 mmol / L ~ 0. 2 mmol / L ~ (2) mmol / L ~ 2 mmol / L ~ 4 mmol / L) and 6 h / 12 h / 24 h ~ 24 h / L ~ 36 h / L ~ 48 h). The appropriate concentration and duration of homocysteine intervention were further divided into groups and capsaicin was given. Then the expression of TRPV1 protein on human vascular endothelial cells was detected by immunofluorescence assay. The expression of TRPV1 protein in each group was detected by Tunel staining. Results (1) homocysteine had a significant inhibitory effect on the proliferation of HUVECs, and the absorptivity decreased from 1.03 鹵0.12) to 0.42 鹵0.05 渭 m, and showed a significant dose-effect relationship (P 0.01), and when the intervention concentration began with 0.5mmol/L, the concentration of homocysteine decreased from 1.03 鹵0.12 to 0.42 鹵0.05G, and the concentration of Hcy decreased from 1.03 鹵0.12 to 0.42 鹵0.05g. After 24 hours of intervention, the proliferation of human umbilical vein endothelial cells was inhibited by P0. 05 and P0. 05) the expression of TRPV1 protein was detected on HUVECs. The expression of TPRV1 was decreased in 0.5 mmol / L 1 mmol / L / L 2 mmol / L group compared with the control group (P 0. 05%), and the apoptosis of the cells was significantly lower than that of the control group (P 0. 05), and the expression of TPRV1 was significantly lower than that of the control group (P < 0. 05) after the intervention of homocysteine. In the exposure group, the concentration of capsaicin was increased by P0.05 (4) capsaicin. Compared with the control group, the expression of TRPV1 protein increased and the apoptosis rate decreased, and there was a significant difference in the expression of TRPV1 protein between the two groups. Conclusion: high homocysteine can promote the apoptosis of HUVECs by down-regulating the expression of TRPV1 protein, and capsaicin can inhibit the apoptosis of endothelial cells induced by high homocysteine.
