【摘要】：目的探究obestatin在心肌梗死后心力衰竭(以下簡(jiǎn)稱心衰)大鼠循環(huán)和腎臟組織中的表達(dá)及其可能的生物學(xué)意義。方法以冠狀動(dòng)脈結(jié)扎法復(fù)制大鼠心肌梗死后心衰模型,酶聯(lián)免疫吸附法檢測(cè)大鼠血漿obestatin濃度,免疫組織化學(xué)法檢測(cè)obestatin在腎臟組織的表達(dá)并分析累積光密度值,比較不同組的差異,Western blot檢測(cè)不同組腎臟組織表達(dá)obestatin水平,并進(jìn)行半定量分析。結(jié)果心衰組大鼠循環(huán)血漿內(nèi)obestatin濃度較正常組及假手術(shù)組增高(P0.05),免疫組織化學(xué)法顯示,obestatin在大鼠腎臟腎小管和集合管表達(dá),心衰組大鼠腎臟組織obestatin表達(dá)較正常組和假手術(shù)組升高(P0.05)。結(jié)論 Obestatin在心力衰竭大鼠循環(huán)和腎臟表達(dá)增高,可能以內(nèi)分泌及自分泌或旁分泌形式參與心衰水潴留的發(fā)生、發(fā)展。
[Abstract]:Objective to investigate the expression of obestatin in circulatory and renal tissues of rats with heart failure after myocardial infarction and its possible biological significance. Methods Rat heart failure model was established by coronary artery ligation, plasma obestatin concentration was detected by enzyme-linked immunosorbent assay (Elisa), expression of obestatin in renal tissue was detected by immunohistochemistry and cumulative optical density was analyzed. The expression of obestatin in renal tissues of different groups was detected by Western blot and semi-quantitative analysis was performed. Results the level of obestatin in circulating plasma in CHF group was higher than that in normal group and sham operation group (P 0.05). Immunohistochemical method showed that OBESTatin was expressed in renal tubules and collecting ducts of rats, and the expression of obestatin in kidney tissue of CHF group was higher than that of normal group and sham operation group (P 0.05). Conclusion the increased expression of Obestatin in circulation and kidney of rats with heart failure may be involved in the occurrence and development of water retention in heart failure in the form of endocrine autocrine or paracrine.
