MGP與主動脈瓣膜鈣化的相關性研究

【摘要】:背景主動脈瓣膜鈣化性疾病(Calcific Aortic Valve Disease,CAVD)的發(fā)病率隨著老齡化的加劇逐年升高,具體機制不清楚。CAVD是調(diào)控機制復雜的瓣膜間質(zhì)細胞成骨樣分化過程。實驗發(fā)現(xiàn),基質(zhì)Gla蛋白(Matrix Gla Protein MGP)與動物主動脈瓣膜鈣化有關,是瓣膜鈣化的一種抑制劑,在血清和瓣膜組織中均有表達。但關于MGP與人主動脈瓣膜的關系尚未見報道。目的檢測MGP在人主動脈瓣膜鈣化中的表達情況,反映MGP與人主動脈瓣膜鈣化的關系。方法1.需行瓣膜置換的無病變的馬方綜合癥及主動脈夾層患者的主動脈瓣膜10例做對照組;單純主動脈瓣膜鈣化需行主動脈置換患者的瓣膜做實驗組,結(jié)合術前超聲及術中探查情況分為中度鈣化組10例,重度鈣化組10例;2.術前30分鐘分別取正常組及中、重度鈣化組患者血標本;3.Elisa法、Western-Blot法測定對照組及實驗組血清及瓣膜組織中MGP水平。結(jié)果主動脈瓣膜MGP的表達與對照組相比,中、重度鈣化組主動脈瓣膜MGP表達水平降低(P0.05);血清MGP蛋白水平與對照組相比,中、重度鈣化組的主動脈瓣膜MGP水平降低(P0.05)。結(jié)論本研究發(fā)現(xiàn),隨著鈣化的加重,血清及瓣膜中MGP表達量逐漸降低,特別是重度鈣化組中MGP表達量明顯降低。說明MGP的表達量隨瓣膜鈣化程度的加重呈降低趨勢。
[Abstract]:Background the incidence of aortic valve calcification disease Calcific Aortic Valve disease is increasing with the aggravation of aging, and the mechanism is not clear. CAVD is the process of osteoblast-like differentiation of valvular interstitial cells with complex regulation mechanism. It was found that matrix Gla Protein MGPs are associated with aortic valve calcification and are an inhibitor of valvular calcification, and are expressed in both serum and valve tissues. However, the relationship between MGP and human aortic valve has not been reported. Aim to detect the expression of MGP in human aortic valve calcification and to reflect the relationship between MGP and human aortic valve calcification. Method 1. The control group consisted of 10 patients with valvular valvular disease and aortic dissection, and 10 patients with aortic valve calcification were treated with aortic valve replacement as experimental group. Combined with preoperative ultrasound and intraoperative exploration, 10 cases were divided into moderate calcification group (n = 10) and severe calcification group (n = 10). Serum MGP levels in control group and experimental group were determined by Western-Blot 30 minutes before operation. Results compared with the control group, the expression of MGP in aortic valve was significantly decreased in moderate calcified group and serum MGP protein level was decreased in moderate calcified group compared with control group, while in severe calcified group, the MGP level of aortic valve was lower than that of control group. Conclusion with the increase of calcification, the expression of MGP in serum and valve decreased gradually, especially in the severe calcification group. The results showed that the expression of MGP decreased with the severity of calcification.
