【摘要】:目的研究心臟發(fā)育相關(guān)基因心肌轉(zhuǎn)錄因子NKX2.5和鋅指轉(zhuǎn)錄因子GATA4在先天性心臟病(CHD)的相互作用關(guān)系。方法構(gòu)建基因NKX2.5和GATA4真核表達(dá)質(zhì)粒及腦鈉肽啟動(dòng)子熒光素酶報(bào)告基因表達(dá)質(zhì)粒,真核表達(dá)質(zhì)粒經(jīng)轉(zhuǎn)染后,利用免疫共沉淀技術(shù)證明基因NKX2.5和GATA4兩者是否有相互作用,通過(guò)熒光素酶分析技術(shù)分析基因NKX2.5和GATA4對(duì)腦鈉肽啟動(dòng)子序是否存在協(xié)同激活作用。結(jié)果 p CMV-HA-NKX2.5與Pre-immune作用后蛋白表達(dá)量0,與Anti-Myc作用后蛋白表達(dá)量為3.63±1.23,與Input作用后蛋白表達(dá)量為2.95±1.05,NKX2.5可將GATA4沉淀下來(lái);p CMV-Myc-GATA4與Pre-immune作用后蛋白表達(dá)量0,與Anti-Myc作用后蛋白表達(dá)量為5.63±2.06,與Input作用后蛋白表達(dá)量為5.75±2.86,GATA4可將NKX2.5沉淀下來(lái);腦鈉肽(BNP)啟動(dòng)子熒光表達(dá)質(zhì)粒與兩者共轉(zhuǎn)染比分別單獨(dú)轉(zhuǎn)染NKX2.5和GATA4對(duì)腦鈉肽啟動(dòng)子的激活作用更強(qiáng)。結(jié)論 NKX2.5和GATA4之間具有相互作用,在心臟發(fā)育過(guò)程中起著重要的協(xié)同作用。
[Abstract]:Objective to study the interaction between cardiac development-related gene NKX2.5 and zinc finger transcription factor (GATA4) in congenital heart disease (CHD).Methods the eukaryotic expression plasmids of gene NKX2.5 and GATA4 and the plasmids expressing luciferase reporter genes of brain natriuretic peptide promoter were constructed. After transfection of eukaryotic expression plasmids, the interaction between NKX2.5 and GATA4 was proved by immunoprecipitation.Luciferase analysis was used to determine whether the gene NKX2.5 and GATA4 had synergistic effects on brain natriuretic peptide promoter sequence.Results the protein expression of p CMV-HA-NKX2.5 and Pre-immune was 0, that of Anti-Myc was 3.63 鹵1.23, that of Input was 2.95 鹵1.05NKX2.5, that of GATA4 precipitated by p CMV-Myc-GATA4 and Pre-immune was 0, and that of Anti-Myc was 0.After treatment with Input, the protein expression was 5.75 鹵2.86 and GATA4 could precipitate NKX2.5.The fluorescent expression plasmid of brain natriuretic peptide (BNP) promoter and co-transfection of NKX2.5 and GATA4 had stronger effect on the activation of brain natriuretic peptide promoter.Conclusion there is an interaction between NKX2.5 and GATA4, which plays an important synergistic role in cardiac development.
