【摘要】：目的正交設計篩選苯和環(huán)磷酰胺聯(lián)合使用建立新西蘭兔再生障礙性貧血模型的適宜劑量。方法新西蘭兔,采用正交試驗設計,應用L9(34)正交表對苯的劑量(A)、環(huán)磷酰胺的劑量(B)、苯的注射次數(shù)(C)、環(huán)磷酰胺的注射次數(shù)(D)四個因素及它們各自不同的3個水平在建立新西蘭兔再生障礙性貧血模型,并最終從9個實驗組中優(yōu)選出建模的較優(yōu)方案。給藥方法為先背部皮下注射苯,隔日1次,再耳緣靜脈注射環(huán)磷酰胺,每天注射,均按照規(guī)定次數(shù)注射。每6 d檢測1次血常規(guī),于建模前、建模后36d取小段股骨進行骨髓組織學檢查,觀察變化。結(jié)果將9組白細胞、紅細胞、血小板計數(shù)進行對比分析,4~9組新西蘭兔再障模型建立成功,第7、8、9組與其他組日均下降速率之差有統(tǒng)計學意義(P0.05),其中第7組骨髓切片顯示骨髓增生低下,造血組織減少,巨核細胞減少或消失,脂肪細胞增多。隨訪發(fā)現(xiàn),第7組骨髓抑制一直存在,與該病臨床特點相符合。結(jié)論第7組采用苯1.5 m L/kg,8次/d,環(huán)磷酰胺10 mg/kg,4次/d,這一組合劑量建立的新西蘭兔AA模型建模周期短、且模型穩(wěn)定,是一種可廣泛應用于動物實驗的建模方法。
[Abstract]:Objective to study the optimal dose of benzene and cyclophosphamide in aplastic anemia model in New Zealand rabbits.Methods New Zealand rabbits were designed by orthogonal test.In order to establish aplastic anemia model of New Zealand rabbits, four factors were used to establish aplastic anemia model in New Zealand rabbits, which were the dose of benzene, the dose of cyclophosphamide, the times of injection of benzene and the times of injection of cyclophosphamide, respectively.Finally, the optimal scheme of modeling was selected from 9 experimental groups.First, benzene was injected subcutaneously into the back, once every other day, and then cyclophosphamide was injected into the ear vein every day.Blood routine was detected once every 6 days, and bone marrow histological examination was performed 36 days after modeling.Results the white blood cell, red blood cell and platelet count in 9 groups were compared and analyzed. The model of aplastic anemia in 9 groups of New Zealand rabbits was established successfully.There was significant difference in the daily average rate of decline between group 7 and other groups (P 0.05). Bone marrow sections in group 7 showed low bone marrow proliferation, decreased hematopoietic tissue, decreased or disappeared megakaryocyte, and increased adipocytes.Follow-up showed that bone marrow suppression existed all the time in group 7, which was consistent with the clinical features of the disease.Conclusion the AA model of New Zealand rabbits established with benzene 1.5 mL / kg / kg 8 times / d and cyclophosphamide 10 mg / kg / d 4 times / d in the seventh group is a stable and stable model. It can be widely used in animal experiments.
