Irbesartan對(duì)冠狀動(dòng)脈粥樣硬化小鼠的治療作用及可能機(jī)制

【摘要】:目的:明確Irbesartan對(duì)冠狀動(dòng)脈粥樣硬化小鼠的治療作用,并進(jìn)一步分析可能的機(jī)制。方法:將16只雄性Apo E-/-小鼠給予高脂飲食(1.25%膽固醇,10%脂肪)構(gòu)建冠心病模型。模型建立后將小鼠隨機(jī)分為治療組[Irbesartan,50mg/(kg·d),4周]及對(duì)照組(等量生理鹽水灌胃對(duì)照)。采用HE、ELISA、Western blot及免疫熒光技術(shù)分析療效及機(jī)制。結(jié)果:治療組小鼠動(dòng)脈粥樣斑塊面積顯著低于對(duì)照組(P0.05)。治療組血管內(nèi)IL-1β、IL-6及TNF-α水平顯著低于對(duì)照組(P0.05)。治療組小鼠血管周圍脂肪組織內(nèi)PPAR-γ及Adiponectin水平顯著升高,而Leptin水平顯著降低,與對(duì)照組相比差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。Western blot及免疫熒光證實(shí),Irbesartan顯著抑制了治療組小鼠血管周圍脂肪組織內(nèi)的NF-κB信號(hào)通路。結(jié)論:Irbesartan通過(guò)調(diào)節(jié)血管周圍脂肪組織內(nèi)PPAR-γ-NF-κb(p65)信號(hào)通路,調(diào)節(jié)血管周圍脂肪組織功能及炎癥,從而發(fā)揮抗動(dòng)脈粥樣硬化療效。
[Abstract]:Objective: to investigate the therapeutic effect of Irbesartan on mice with coronary atherosclerosis. Methods: 16 male Apo E-r-mice were given 1.25% cholesterol 10% fat in a high-fat diet to establish a coronary heart disease model. After the establishment of the model, the mice were randomly divided into two groups: treatment group [Irbesartanan 50mg / kg / kg / d + 4 weeks] and control group (et al.). The therapeutic effect and mechanism were analyzed by Western blot and immunofluorescence technique. Results: the area of atherosclerotic plaques in the treatment group was significantly lower than that in the control group (P 0.05). The levels of IL-1 尾 -IL-6 and TNF- 偽 in the treatment group were significantly lower than those in the control group. The levels of PPAR- 緯 and Adiponectin in perivascular adipose tissue of mice in the treatment group were significantly higher than those in the control group. However, the level of Leptin decreased significantly, The results showed that Irbesartan significantly inhibited NF- 魏 B signaling pathway in perivascular adipose tissue of mice in the treatment group by regulating the PPAR- 緯 -NF- 魏 p65 signal pathway in perivascular adipose tissue. Regulate the function of perivascular adipose tissue and inflammation, thus exerting the effect of anti-atherosclerosis.
