運(yùn)動(dòng)訓(xùn)練對(duì)高血壓心臟功能的影響及其機(jī)制

【摘要】:目的:探討運(yùn)動(dòng)訓(xùn)練對(duì)高血壓大鼠的血壓、心功能、心肌細(xì)胞收縮性、心肌細(xì)胞肌絲鈣敏感性的影響。方法:大鼠隨機(jī)分為四組,分別為高血壓組,高血壓運(yùn)動(dòng)組,假手術(shù)組,假手術(shù)運(yùn)動(dòng)組。利用血管緊張素II誘導(dǎo)高血壓。多動(dòng)物多通道尾袖套法無創(chuàng)血壓系統(tǒng),監(jiān)測大鼠血壓。心臟超聲儀測左室射血分?jǐn)?shù)。分離的心肌細(xì)胞,采用IonOptix單細(xì)胞收縮檢測系統(tǒng)檢測心肌細(xì)胞收縮性及心肌細(xì)胞肌絲鈣敏感性。結(jié)果:(1)血壓:運(yùn)動(dòng)2周,高血壓組收縮壓與高血壓運(yùn)動(dòng)組收縮壓有統(tǒng)計(jì)學(xué)意義(P0.05),高血壓運(yùn)動(dòng)組的收縮壓明顯得到控制。假手術(shù)組收縮壓與假手術(shù)運(yùn)動(dòng)組收縮壓間無統(tǒng)計(jì)學(xué)意義(P0.05)。高血壓組舒張壓與高血壓運(yùn)動(dòng)組舒張壓間有統(tǒng)計(jì)學(xué)意義(P0.05),高血壓運(yùn)動(dòng)組的舒張壓明顯得到控制。假手術(shù)組舒張壓與假手術(shù)運(yùn)動(dòng)組舒張壓間無統(tǒng)計(jì)學(xué)意義(P0.05);(2)左室射血分?jǐn)?shù)(LVEF):運(yùn)動(dòng)訓(xùn)練后,高血壓組LVEF與高血壓運(yùn)動(dòng)組LVEF間有統(tǒng)計(jì)學(xué)意義(P0.05)。運(yùn)動(dòng)訓(xùn)練后大鼠LVEF有顯著提高;(3)心肌細(xì)胞收縮性:運(yùn)動(dòng)訓(xùn)練后,高血壓組心肌細(xì)胞收縮性與高血壓運(yùn)動(dòng)組心肌細(xì)胞收縮性間有統(tǒng)計(jì)學(xué)意義(P0.05),運(yùn)動(dòng)訓(xùn)練后大鼠心肌收縮性明顯恢復(fù);(4)肌絲鈣敏感性:運(yùn)動(dòng)訓(xùn)練使,高血壓運(yùn)動(dòng)組肌節(jié)縮短長度與Ca2+濃度關(guān)系曲線向左移;運(yùn)動(dòng)訓(xùn)練后高血壓組EC50與高血壓運(yùn)動(dòng)組EC50間有統(tǒng)計(jì)學(xué)意義,(P<0.05),運(yùn)動(dòng)訓(xùn)練使EC50縮短。結(jié)論:1.運(yùn)動(dòng)訓(xùn)練能有效控制高血壓大鼠的收縮壓和舒張壓;2.運(yùn)動(dòng)訓(xùn)練通過恢復(fù)心肌細(xì)胞肌絲Ca2+敏感性達(dá)到對(duì)心肌細(xì)胞收縮性及心功能的改善。
[Abstract]:Objective: to investigate the effects of exercise training on blood pressure, cardiac function, myocardial contractility and myofilm sensitivity in hypertensive rats. Methods: rats were randomly divided into four groups: hypertension group, hypertension exercise group and sham operation group. Sham operation exercise group. Hypertension was induced by angiotensin II. Non-invasive blood pressure system with multi-animal multi-channel tail sleeve method was used to monitor blood pressure in rats. Left ventricular ejection fraction (LVEF) was measured by cardiac ultrasound. Isolated cardiomyocytes were isolated. IonOptix single cell contraction detection system was used to detect the contractility of cardiomyocytes and the sensitivity of myofilm calcium. Results: blood pressure: exercise for 2 weeks. Systolic blood pressure in hypertension group was significantly higher than that in exercise group (P 0.05). Systolic blood pressure in exercise group was significantly controlled. There was no significant difference between systolic blood pressure in sham operation group and exercise group (P 0.05). There was significant difference between diastolic blood pressure (DBP) and diastolic blood pressure (DBP) in hypertensive exercise group (P 0.05). Diastolic blood pressure (DBP) in exercise group was significantly controlled. There was no significant difference between diastolic blood pressure (DBP) and diastolic blood pressure (DBP) in sham-operated exercise group (P < 0.05); left ventricular ejection fraction (LVEFV): after exercise training, There was significant difference between LVEF in hypertension group and LVEF in hypertensive exercise group (P 0.05). The contractility of cardiac myocytes in rats after exercise training was significantly increased after exercise training (P < 0.05). The contractility of cardiac myocytes in hypertension group was significantly different from that in exercise group (P 0.05). The contractility of myocardium recovered significantly after exercise training (P < 0.05). The relationship between the length of muscle ganglion shortening and the concentration of Ca2 in hypertensive exercise group was shifted to the left. After exercise training, there was significant difference between EC50 and EC50 in hypertension group (P < 0.05), and EC50 was shortened by exercise training. Conclusion 1. Exercise training can effectively control systolic and diastolic blood pressure in hypertensive rats. The sensitivity of myofilament Ca2 to cardiac contractility and cardiac function was improved.
