文章摘要:目的:探討銀杏葉提取物對野百合堿所致肺動(dòng)脈高壓(PAH)的早期保護(hù)作用。方法:30只雄性 SD大鼠隨機(jī)分為3組:對照組、PAH 模型組和治療組,每組10只。PAH 模型組和治療組大鼠采用脊背部皮下注射1%野百合堿60 mg/kg的方法復(fù)制PAH 模型,注射后第2天開始每日予2 ml 0.9%氯化鈉注射液(PAH 模型組)或60 mg/kg銀杏葉提取物(治療組)灌胃;對照組脊背部皮下注射等量溶劑,注射后第2天開始每日予2 ml 0.9%氯化鈉注射液灌胃。第22天采用頸外靜脈右心導(dǎo)管法測定平均肺動(dòng)脈壓(mPAP)、右心室收縮壓(RVSP),測定右心室、左心室、室間隔的重量,計(jì)算右心室肥厚指數(shù)(RVHI);采用 HE染色觀察肺細(xì)小動(dòng)脈組織結(jié)構(gòu)情況,并計(jì)算直徑50~150μm的肺小動(dòng)脈血管管壁厚度占血管厚度百分比(WA%)和血管管壁面積與血管面積比值(WV%);采用免疫組化方法觀察內(nèi)皮型一氧化氮合成酶(eNOS)及內(nèi)皮素-1(ET-1)在大鼠肺組織中的表達(dá)情況。結(jié)果:PAH 模型組、治療組的 mPAP、RVSP、RVHI、WA%、WV%與對照組比較均明顯升高(P〈0.05),治療組的 mPAP、RVSP、WA%、WV%較 PAH 模型組均降低(P〈0.05),而治療組與 PAH 模型組的RVHI差異無顯著性。免疫組化結(jié)果顯示,3組 eNOS表達(dá),對照組〉治療組〉PAH 模型組;3組 ET-1的表達(dá), PAH 模型組〉治療組〉對照組。結(jié)論:銀杏葉提取物通過抑制 ET-1的合成、減輕內(nèi)皮細(xì)胞損傷、維持 eNOS的表達(dá),達(dá)到減緩PAH 進(jìn)展的作用。
Abstr:Objective:To investigate the protective effect of ginkgo biloba extract (GBE)on monocrotaline-in-duced pulmonary hypertension in early stage.Methods:Thirty male SD rats were randomly divided into three groups :control group,pulmonary hypertension model group (model group),ginkgo biloba extract treatment group (treatment group,n=10). The pulmonary hypertension model was reproduced by subcutaneous injection of monocrotaline (60 mg/kg)on the back,and then,2 ml normal saline or 60 mg/kg GBE was given by gavage from the second day in model group or treatment group. In control group,the same volume of vehicle was subcutane-ously inj ected instead of monocrotaline,and 2 ml normal saline was given by gavage from the second day. Mean pulmonary arterial pressure (mPAP)and right ventricular systolic pressure (RVSP)were measured through cath-eterization into the right ventricle via the right jugular vein on the 22nd day. The right ventricular,left ventricle, and interventricular septum were weighed to calculate the right ventricular hypertrophic index (RVHI).The structure of pulmonary arterioles was observed with HE staining. The ratio of wall thickness/diameter of arteri-oles (WA%),and the ratio of vascular wall area/vascular cross-sectional area (WV%)of pulmonary arterioles with diameter of 50-150μm were calculated. The expression of endothelial nitric oxide synthase (eNOS)and en-dothelin-1 (ET-1)in the lung tissue of rats were assayed by immunohistochemical method.Results:Compared to the control group,mPAP,RVSP,RVHI,WA% and WV% were significantly increased in model group and treatment group (P〈0.05).In the treatment group,mPAP,RVSP,WA% and WV% were lower than those of the model group (P〈0.05),but no significant change was found in RVHI.
