【摘要】：[目的]檢測我科血液腫瘤患者中WNK1-WAC基因易位突變的發(fā)生率和分布情況,結(jié)合臨床資料試圖分析其在ANLL及其他血液腫瘤的致病及復(fù)發(fā)機(jī)理。[方法]收集2014年4月-2015年5月在昆明醫(yī)科大學(xué)第一附屬醫(yī)院血液內(nèi)科收治住院的300例不同類型血液腫瘤患者的骨髓液樣品及10例非惡性血液腫瘤患者骨髓液樣品(作對(duì)照)。采用PCR、RT-PCR結(jié)合瓊脂糖凝膠電泳法檢測血液腫瘤患者中WNK1-WAC基因易位突變并分析。[結(jié)果]300例不同血液腫瘤患者(141例AML、32例ALL、30例CML、14例CLL、18例MDS、25例MM、40例NHL)及10例非惡性血液腫瘤患者中均未檢測到WNK1-WAC基因易位突變。[結(jié)論]在我科血液腫瘤患者中,未檢測到WNK1-WAC基因易位突變,可能是由于樣本量小,或在該人種中,無WNK1-WAC基因易位突變,疾病的發(fā)生發(fā)展與該基因無關(guān)。
[Abstract]:[objective] to detect the incidence and distribution of WNK1-WAC gene translocation mutation in patients with hematologic tumors in our department. Combined with clinical data, this paper attempts to analyze the pathogenesis and recurrence mechanism of ANLL and other blood tumors. [methods] 300 cases of different types of patients admitted to the Department of Hematology, first affiliated Hospital of Kunming Medical University from April 2014 to May 2015 were collected. Bone marrow fluid samples from patients with hematologic tumors and bone marrow fluid samples from 10 patients with non-malignant hematologic tumors were used to detect translocation mutations of WNK1-WAC gene in patients with hematologic tumors by RT-PCR and agarose gel electrophoresis. WNK1-WAC gene translocation mutations were not detected in 141 patients with different blood tumors, 30 patients with WNK1-WAC, 30 patients with CML, 14 patients with WNK1-WAC, 18 patients with MDS, 25 patients with MML and 40 patients with non-malignant hematological tumors. [conclusion] in our department, no translocation mutation of WNK1-WAC gene was detected in patients with hematologic tumors. No translocation mutation of WNK1-WAC gene was detected, which may be due to the small sample size or the absence of translocation mutation of WNK1-WAC gene in this population, and the occurrence and development of the disease have nothing to do with the gene.
