胰島素及葡萄糖對(duì)SD大鼠內(nèi)皮細(xì)胞表達(dá)sES以及平滑肌細(xì)胞表達(dá)MMP-9、TIMP-1的影響

【摘要】:目的:研究胰島素與葡萄糖對(duì)SD大鼠內(nèi)皮細(xì)胞表達(dá)可溶性E選擇素(sES)及平滑肌細(xì)胞表達(dá)基質(zhì)金屬蛋白酶9(MMP-9)、組織抑制因子-1(TIMP-1)的影響。方法:培養(yǎng)SD大鼠主動(dòng)脈內(nèi)皮細(xì)胞及頸內(nèi)動(dòng)脈平滑肌細(xì)胞,用不同濃度的胰島素和葡萄糖對(duì)其進(jìn)行刺激,以酶聯(lián)免疫吸附試驗(yàn)(ELISA)分別測(cè)定上清液sES、MMP-9及TIMP-1的水平。結(jié)果:與對(duì)照組(無(wú)胰島素)相比,胰島素刺激組sES和MMP-9表達(dá)水平顯著增高(P0.05和P0.05),TIMP-1表達(dá)則顯著降低(P0.05),且差異有統(tǒng)計(jì)學(xué)意義。經(jīng)Pearson相關(guān)分析顯示,胰島素濃度與sES(r=0.841,P0.001)及MMP-9(r=0.815,P0.001)呈正相關(guān),與TIMP-1(r=--0.963,P0.001)呈負(fù)相關(guān)。與對(duì)照組(無(wú)葡萄糖)相比,葡萄糖刺激組sES和MMP-9表達(dá)水平顯著增高(P0.005和P0.001),TIMP-1表達(dá)則顯著降低(P0.001),且差異有統(tǒng)計(jì)學(xué)意義。經(jīng)Pearson相關(guān)分析顯示,葡萄糖濃度與sES(r=0.972,P0.001)及MMP-9(r=0.973,P0.001)呈正相關(guān),與TIMP-1(r=-0.963,P0.001)呈負(fù)相關(guān)。結(jié)論:胰島素及葡萄糖在一定濃度范圍內(nèi)能促進(jìn)SD大鼠內(nèi)皮細(xì)胞sES及平滑肌細(xì)胞MMP-9表達(dá),抑制平滑肌細(xì)胞TIMP-1表達(dá)。
[Abstract]:Objective: to study the effects of insulin and glucose on the expression of soluble E selectin ESSS (soluble E selectin ESS) and matrix metalloproteinase-9 (MMP-9), tissue inhibitor of metalloproteinase-1 (TIMP-1) in rat endothelial cells. Methods: aortic endothelium of SD rats was cultured in vitro. Cells and smooth muscle cells of the internal carotid artery, The levels of sESU MMP-9 and TIMP-1 in supernatant were determined by enzyme linked immunosorbent assay (Elisa) with different concentrations of insulin and glucose. In insulin-stimulated group, the expression of sES and MMP-9 increased significantly (P0.05) and the expression of TIMP-1 decreased significantly (P 0.05), and the difference was statistically significant. The Pearson correlation analysis showed that the insulin concentration was positively correlated with the expression of sESnr ~ (0.841) P _ (0.001) and MMP-9 ~ (9) (P ~ (0.815) P _ (0.001)). The expression of sES and MMP-9 in glucose stimulated group was significantly higher than that in control group (P 0. 005 and P 0. 001), and the expression of TIMP-1 was significantly lower than that in control group (P 0. 005 and P 0. 001 P 0. 001). The Pearson correlation analysis showed that the expression of sES and MMP-9 in glucose stimulated group was significantly lower than that in control group (P 0. 005 and P 0. 001). There was a positive correlation between glucose concentration and the expression of sES and MMP-9 in the endothelial cells and smooth muscle cells of SD rats, and a negative correlation between the concentration of glucose and TIMP-1 / 0.963P 0.001. Conclusion: insulin and glucose can promote the expression of sES and MMP-9 in smooth muscle cells and inhibit the expression of TIMP-1 in smooth muscle cells of SD rats.
