【摘要】：目的通過體外培養(yǎng)人主動脈平滑肌細(xì)胞(HASMCs),研究胃促生長素(ghrelin)對血管平滑肌細(xì)胞(VSMC)增殖及線粒體融合蛋白2(Mfn-2)表達(dá)的影響。方法體外培養(yǎng)HASMCs,第4~6代細(xì)胞用于試驗(yàn)。給予不同濃度(10~(-9)、10~(-8)、10~(-7)、10~(-6)、10~(-5) mol/L)的ghrelin或10~(-6) mol/L ghrelin不同時(shí)間(0、6、12、18、24h)處理,用四甲基偶氮唑藍(lán)比色(MMT)法觀察其對HASMC增殖的影響。RT-PCR,Western blot方法檢測不同處理方法對Mfn-2表達(dá)的影響。結(jié)果 10~(-7)~10~(-5) mol/L的ghrelin可明顯抑制HASMC增殖,濃度為10~(-6) mol/L抑制作用最為明顯(P0.01)。ghrelin在6~24h內(nèi)均能明顯抑制HASMC增殖,在24h達(dá)最高峰(P0.01)。10~(-6) mol/L ghrelin能明顯上調(diào)Mfn-2 mRNA和蛋白的表達(dá)(P0.01)。10~(-6)mol/L ghrelin在18h上調(diào)Mfn-2mRNA和蛋白表達(dá)的作用最為明顯(P0.01)。結(jié)論 ghrelin可能通過上調(diào)Mfn-2的表達(dá)來抑制HASMC增殖。
[Abstract]:Objective to study the effects of ghrelin on the proliferation of vascular smooth muscle cells (VSMCs) and the expression of mitochondrial fusion protein 2Mfn-2 in cultured human aortic smooth muscle cells (HASMCs) in vitro. Methods HASMCswere cultured in vitro, and the 4th generation of HASMCswere used to test the effects of ghrelin on the proliferation of vascular smooth muscle cells (VSMCs) and the expression of mitochondrial fusion protein 2Mfn-2. The inhibitory effect of mol/L on HASMC proliferation was most obvious at the concentration of 10g / L -6) P0.01C. Ghrelin could significantly inhibit the proliferation of HASMC within 6h after treatment. At 24 h, the expression of Mfn-2mRNA and protein could be upregulated by P0.01L / L ghrelin. Ghrelin may inhibit the proliferation of HASMC by upregulating the expression of Mfn-2 and P0.01mol / L ghrelin at 18h. Conclusion the expression of ghrelin may inhibit the proliferation of HASMC by upregulating the expression of Mfn-2.
