丙型肝炎病毒NS2TP基因調(diào)節(jié)機(jī)制的研究

目的:NS2TP基因是抑制性消減雜交技術(shù)(SSH)篩選出的HCV NS2反式激活的基因之一。本實(shí)驗(yàn)中,我們對HCV NS2反式激活NS2TP基因從翻譯水平和轉(zhuǎn)錄水平進(jìn)行驗(yàn)證,進(jìn)而定位NS2TP基因啟動(dòng)子的核心區(qū)域,并初步探究了NS2TP基因的轉(zhuǎn)錄調(diào)節(jié)機(jī)制。方法:(1) HCV NS2對NS2TP基因反式激活作用的驗(yàn)證。首先,運(yùn)用pcDNA3.1(–)-NS2表達(dá)載體轉(zhuǎn)染HepG2細(xì)胞,以pcDNA3.1(–)為陰性對照組。提取總蛋白、總RNA,然后分別從翻譯水平,即Western Blot實(shí)驗(yàn),和轉(zhuǎn)錄水平,即進(jìn)行Real-time PCR實(shí)驗(yàn)。分析和研究NS2對NS2TP基因的反式激活作用。(2) NS2TP基因啟動(dòng)子的活性分析與核心區(qū)的確定。首先,成功構(gòu)建pGL4.10-NS2TP啟動(dòng)子雙熒光素酶報(bào)告基因表達(dá)載體系統(tǒng),運(yùn)用構(gòu)建的不同截短型載體轉(zhuǎn)染HepG2細(xì)胞,在對啟動(dòng)子活性進(jìn)行分析的基礎(chǔ)上,實(shí)驗(yàn)確定了NS2TP基因啟動(dòng)子活性的核心區(qū)域,同時(shí)也分析了NS2對NS2TP基因啟動(dòng)子的調(diào)節(jié)作用。(3) NS2TP基因轉(zhuǎn)錄調(diào)節(jié)因子CREB作用啟動(dòng)子核心區(qū)的驗(yàn)證。運(yùn)用生物信息學(xué)技術(shù)分析了啟動(dòng)子...

Real Time PCR 擴(kuò)增反應(yīng)采用了如圖 2-3 顯示的 SYBR Green I 嵌合熒光法,即在反應(yīng)體系中加入與雙鏈 DNA 結(jié)合便可發(fā)出熒光的熒光染料 SYBR Green I,通過檢測 PCR 反應(yīng)液中的熒光信號強(qiáng)度,可以對目的基因進(jìn)行準(zhǔn)確定量,同時(shí)還可以測定擴(kuò)增的目的 DNA 片段的融解溫度。
